Haloperidol Induced Cell Cycle Arrest and Apoptosis in Glioblastoma Cells

Although several antipsychotic drugs have been shown to possess anticancer activities, haloperidol, a first-generation antipsychotic drug, has not been extensively evaluated for potential antineoplastic properties. The aim of this study was to investigate the antitumoral effects of haloperidol in glioblastoma (GBM) U87, U251 and T98 cell lines, and the effects of combined treatment with temozolomide (TMZ) and/or radiotherapy, using 4 Gy of irradiation. The viability and proliferation of the cells were evaluated with trypan blue exclusion assay and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Apoptosis, using the annexin-propidium iodide (PI), and cell cycle, cluster of differentiation (CD) expression and caspase-8 activation were measured using flow cytometry. Treatment with haloperidol significantly reduced cell viability in U87, U251 and T98 GBM cell lines. Haloperidol induced apoptosis in a dose-dependent manner, inhibited cell migration and produced an alteration in the expression of CD24/CD44. The additional effect of haloperidol, combined with temozolomide and radiation therapy, increased tumor cell death. Haloperidol was observed to induce apoptosis and to increase caspase-8 activation. In conclusion, haloperidol may represent an innovative strategy for the treatment of GBM and further studies are warranted in glioma xenograft models and other malignancies.

Circular RNAs (circRNAs) have been shown to modulate gene expression and participate in the development of multiple malignancies. The purpose of this study was to investigate the role of circ_0008039 in breast cancer (BC). The expression of circ_0008039, miR-140-3p, and spindle and kinetochore-associated protein 2 (SKA2) was detected by qRT-PCR. Cell viability, colony formation, migration, and invasion were evaluated using methylthiazolyldiphenyl-tetrazolium bromide (MTT) assay, colony formation assay, and transwell assay, respectively. Glucose consumption and lactate production were measured using commercial kits. Protein levels of hexokinase II (HK2) and SKA2 were determined by western blot. The interaction between miR-140-3p and circ_0008039 or SKA2 was verified by dual-luciferase reporter assay. Finally, a mouse xenograft model was established to investigate the roles of circ_0008039 in BC in vivo. We found that circ_0008039 and SKA2 were upregulated in BC tissues and cells, while miR-140-3p was downregulated. Knockdown of circ_0008039 suppressed BC cell proliferation, migration, invasion, and glycolysis. Moreover, miR-140-3p could bind to circ_0008039 and its inhibition reversed the inhibitory effect of circ_0008039 interference on proliferation, migration, invasion, and glycolysis in BC cells. SKA2 was verified as a direct target of miR-140-3p and its overexpression partially inhibited the suppressive effect of miR-140-3p restoration in BC cells. Additionally, circ_0008039 positively regulated SKA2 expression by sponging miR-140-3p. Consistently, silencing circ_0008039 restrained tumor growth via increasing miR-140-3p and decreasing SKA2. In conclusion, circ_0008039 downregulation suppressed BC cell proliferation, migration, invasion, and glycolysis partially through regulating the miR-140-3p/SKA2 axis, providing an important theoretical basis for treatment of BC.

Effect of oxoanions on oxidant decay, bromate and brominated disinfection by-product formation during chlorination in the presence of copper corrosion products

The present study investigated the effect of oxoanions on catalytic behaviour of copper corrosion products (CCPs) during chlorination of bromide-containing waters. Three types of oxoanions (carbonate, sulphate, and phosphate) and four types of CCPs (Cu2+, Cu(OH)(2), Cu2O, and CuO) were involved in investigation and the effect of oxoanions concentration was also examined. The result indicated that carbonate and sulphate slightly inhibited oxidant decay in the presence of CCPs, but the formation of brominated disinfection by-products (Br-DBPs) remained largely unchanged. In contrast, the presence of phosphate (0.2-1 mM) almost eliminated the catalytic effect of Cu2+. For CCP solids (i.e. Cu(OH)2, Cu2O, and CuO), phosphate preferentially inhibited the formation of bromate rather than Br-DBPs. Despite the catalysis by CCP solids was reduced to some extent, the oxidant decay rate and bromate and Br-DBP formation were still significantly higher than blank groups, even at high phosphate concentration. By testing different addition scheme (simultaneous/sequential addition), it was proposed that phosphate was a strong competitor for hypohalites, rapidly destroying CCPs-hypohalites complexes on some adsorption sites. However, there were some specific sites that can only be adsorbed by hypohalites, leading to the incomplete inhibition of phosphate. Finally, the inhibition effect of phosphate on CCPs catalysis was tested in real water matrix. For Cu2+, higher reduction of bromate and Br-DBPs was found in raw water rather than filtered water, while converse pattern was true for Cu(OH)(2) and Cu2O, and this discrepancy can be ascribed to the difference in catalytic mechanism between Cu2+ and CCP solids. A simple hypochlorous acid signaling probe based on resorufin carbonodithioate and its biological application

A novel hypochlorous-acid-selective signaling probe based on the carbonodithioate derivative of resorufin (RT-1) was developed. Probe RT-1 showed prominent colorimetric and turn-on type fluorescence signaling behavior exclusively toward hypochlorous acid, induced by oxidative hydrolysis, to regenerate resorufin dye. Hypochlorous acid signaling was not affected by the presence of common metal ions and anions as background, except for the redox active bromide and iodide anions. The detection limit of RT-1 for hypochlorous acid was found to be 2.18 x 10(-9) M (0.11 ppb), and the signaling was completed within 3 min. This was rationalized by density functional theory calculations, which demonstrated that the CS sulfur atom of the former is more negatively charged than that of the latter. Enhanced toughness and strength of boron carbide ceramics with reduced graphene oxide fabricated by hot pressing

Boron carbide (B4C) hybrids with different contents of graphene oxide (GO) were prepared by a heterogeneous co-precipitation method using cetyltrimethyl ammonium bromide (CTAB) as the cationic surfactant. The asobtained mixtures were further hot-pressed at 1950 degrees C for 60 min under 30 MPa, by which B4C-reduced GO (rGO) composites were fabricated. It was found that the addition of only 0.5 wt% rGO could alter the predominance of trans-granular fracture in monolithic B4C ceramic material to mixed trans-granular and intergranular modes in B4C-rGO composites. The flexural strength and fracture toughness of the B4C-2 wt% rGO were increased by 31% (from 350 to 455 MPa) and 83% (from 3.20 to 5.85 MPa.m(1/2)), respectively, compared with those of pure B4C. The improved mechanical properties are attributed to the mechanisms of pull-out and bridging of rGO and crack deflection, as evidenced by microstructural observations. The energy dissipation in the present B4C-rGO composites was further verified using two micromechanical models.

Thermal properties of tetragonal tetra-n-butyl ammonium bromide plus tetra-n-butyl ammonium chloride mixed semiclathrate hydrates based on hydration numbers and guest mole fraction rates

In this study, we investigated variations in the thermal properties of tetragonal tetra-n-butyl ammonium bromide (TBAB)+tetra-n-butyl ammonium chloride (TBAC) mixed semiclathrate hydrates by ion chromatography and differential scanning calorimetry; these variations were assessed both in terms of their hydration numbers and the guest mole fraction rates of the hydrates. TBAB mole fraction rates in mixed hydrate crystals (y(TBAB)(h)) are lower than those in mixed aqueous solutions; therefore, as Cl- has a smaller size difference to a water molecule, it is more easily incorporated into the host frameworks of hydrates than Br-. The melting points of mixed hydrates were observed at 286-289K, and the maxima are around y(TBAB)(h)=0.1-0.3 in each hydration number. The increase in melting points largely depends on decreasing y(TBAB)(h) rather than increasing hydration numbers. It is suggested that the combinations of TBAB and TBAC optimize the crystal structures, and melting points of TBAB+TBAC mixed hydrates become anomalously higher than those of the pure TBAC hydrates. Dissociation enthalpies per mass increase from 193Jg(-1) to 215Jg(-1), primarily with decreasing y(TBAB)(h), whereas those per guest moles increase from 154kJmol(-1) to 188kJmol(-1), primarily with increasing hydration numbers rather than y(TBAB)(h).

Kinetic study of semiclathrate hydrates formed with CO2 in the presence of tetra-n-butyl ammonium bromide and tetra-n-butyl phosphonium bromide

The kinetics of semiclathrate hydrates formation with CO2 in the presence of tetra-n-butyl ammonium bromide (TBAB), tetra-n-butyl phosphonium bromide (TBPB), and TBAB + TBPB were investigated. The experiments were conducted at the stoichiometric concentration of TBAB hydrate (2.57 mol%) and three subcoolings (DT = 6 K, 9 K, and 12 K) with the initial pressure fixed at 2.8 MPa. It was found that adding TBPB into the TBAB solution can promote hydrate nucleation, and this promotion effect at a lower subcooling (DT = 6 K) was stronger compared to that at higher subcoolings (DT = 9 K and 12 K). The gas consumption obtained in TBAB, TBPB, and TBAB + TBPB solutions decreased with the increase of sub cooling, despite the increase of the rate of hydrate growth. At a given subcooling, gas consumption (CO2 uptake) in TBAB + TBPB solutions was greater than that in TBAB and TBPB solutions, indicating that hydrate formation kinetics and the CO2 storage capacity were improved in the TBAB + TBPB solution. The largest gas consumption was obtained at DT = 6 K in the TBAB + TBPB solution, which was 28.7% and 17.2% larger than those obtained in TBAB and TBPB solutions, respectively. Therefore, a mixture of TBAB + TBPB is a promising option to improve the kinetics of semiclathrate hydrate formation with CO2. (c) 2020 Elsevier Ltd. All rights reserved.

Folate-Targeted mRNA Delivery Using Chitosan-Functionalized Selenium Nanoparticles: Potential in Cancer Immunotherapy

Systemic messenger RNA (mRNA) delivery, although still in its infancy, holds immense potential for application in cancer vaccination and immunotherapy. Its advantages over DNA transfection make it attractive in applications where transient expression is desired. However, this has proved challenging due to mRNA’s instability and susceptibility to degradation. Selenium is important for immune function and modulation, with selenium nanoparticles (SeNPs) finding a niche in biomedicine as drug delivery vehicles, owing to their biocompatibility, low toxicity, and biodegradability. In this investigation, we synthesized chitosan-coated SeNPs with a folic acid targeting moiety for Fluc mRNA delivery to cancer cells in vitro. Synthesized SeNPs were stable and well dispersed, and ranged from 59 to 102 nm in size. Nanoparticles bound and protected mRNA from RNase degradation, while exhibiting low cytotoxicity in the human embryonic kidney (HEK293), breast adenocarcinoma (MCF-7), and nasopharyngeal (KB) cells in culture. Moderate cytotoxicity evidenced in the colorectal carcinoma (Caco-2) and colon carcinoma (HT-29) cells was attributed to apoptosis induction by selenium, as confirmed by acridine orange/ethidium bromide staining. Selenium uptake studies corroborated the transfection results, where significant transgene expression was evident for the overexpressed folate receptor-positive KB cells when compared to the other cells with less or no folate receptors.
