Augmented therapeutic efficacy of 5-fluorouracil in conjunction with lantibiotic nisin against skin cancer

Chemotherapy, a gold standard for treating most of the cancers, involves drastic side-effects and multidrug resistance. An attractive alternative is development of combination therapy employing antimicrobial peptides with chemotherapeutic drugs. In vivo studies: Anti-cancer therapeutic efficacy of 5-fluororuacil (5-FU) in conjunction with nisin (50 mg/kg body weight) was evaluated against murine skin cancer, in terms of tumor biostatistics, histopathology, electron microscopy, infrared spectroscopy and transcriptional studies. In vitro studies: Dose and time dependent cytotoxicity of agents were assessed against A431 cell line using MIT assay, LDH assay and acridine orange/ethidium bromide dual staining. Significant percentage decrease(s) in mean tumor volume and tumor burden were observed in nisin+ 5-FU combination treated groups as compared to alone treated groups. Histoarchitecture of treated skins demonstrated restoration towards normal skin tissue (being highest in the combination group). Modulation of apoptotic, angiogenic and proliferative genes were observed in treated groups. IC 5 0 of combination was found to be 2 mu g/ml as compared to nisin alone (32 mu g/ml) and 5-FU alone (16 mu g/ml) with combination index of 0.188. Dual staining showed that rate of induction of apoptosis was higher in the combination group as compared to single agents. Investigation of the mechanical properties of Ni-B/hBN composite coatings electrodeposited in presence of CTAB as the surfactant

The effect of hexadecyltrimethylammonium bromide (CTAB) concentration on the galvanostatic electrodeposition of Ni-B matrix nano hexagonal boron nitride (hBN) reinforced composite coatings was investigated. XRD and FE-SEM analyzes were used for crystallographical and morphological investigations, respectively. Vickers indentation, nanoindentation, and nanoscratch experiments were carried out to investigate the micromechanical, nanomechanical and tribological properties, respectively. Potentiodynamic polarization and electrochemical impedance spectroscopy techniques were applied for corrosion characterization. The investigated properties have been improved with the increasing CTAB concentrations up to 900 mu M. In a general manner, CTAB has a beneficial effect at lower concentrations and can be useful for metal matrix nano ceramic reinforced composite coating electrodeposition in the lower concentration range.

microRNA-25 promotes cardiomyocytes proliferation and migration via targeting Bim

microRNAs (miRNAs) are pleiotropic players in cardiac development. Recent evidence have suggested miRNAs as promisingly therapeutic targets for cardiac regeneration. This study aimed to reveal the potential effects of miR-25 on cardiomyocytes proliferation and migration. Sprague-Dawley rats received left coronary occlusion surgery to induce an in vivo model of myocardial ischemia/reperfusion (I/R) injury. Expression changes of miR-25 and Bim were tested by quantitative reverse transcription-polymerase chain reaction (qRT-PCR) and western blot. Besides, primary neonatal and adult cardiomyocytes were transfected by the antisense oligonucleotides or mimic specific for miR-25, and then 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT), 5-ethynyl-2′-deoxyuridine (EdU), Boyden chamber, and terminal-deoxynucleotidyl transferase-mediated nick end labeling (TUNEL) assay were respectively used to determine cardiomyocytes growth and migration. Binding effects of miR-25 on the 3′-untranslated region (3′-UTR) of Bim was assessed by dual-luciferase reporter assay. We found that miR-25 was low expressed, whereas Bim was highly expressed in I/R injury model and hypoxia-stimulated cardiomyocytes. Downregulation of miR-25 in neonatal and adult cardiomyocytes markedly reduced cell proliferation and migration, but promoted apoptosis. Consistently, downregulation of miR-25 decreased the expression of cyclin E2, cyclin D1, and CDK4, and increased the expression of p57 (KIP2) in cardiomyocytes. We additionally found that Bim was a target of miR-25. The inhibitory effects of miR-25 downregulation on cardiomyocytes survival and migration were all significantly attenuated when Bim was silenced. To sum up, our study demonstrates that miR-25 downregulation inhibits cardiomyocytes proliferation and migration, but promotes apoptosis. The role of miR-25 in cardiomyocytes was by targeting Bim.

Regulatory effects of cervical sympathetic trunk and renal sympathetic nerve activities on cerebral blood flow during head-down postural rotations

This study attempts to clarify the neural control of cerebral blood flow (CBF) during head-down postural rotation, which induces a cephalad fluid shift in urethane-anesthetized rats. The animals were placed on a table, tilted to a 45 degrees head-down position over 5 s and maintained in that position. Head-down rotation (HDR) induced a transient decrease (8 +/- 3 mm Hg; mean +/- SE) in mean arterial blood pressure (ABP) at 7.3 +/- 0.3 s after the onset of HDR. The pressure returned to the pre-HDR level within 1 min in the head-down position. Pretreatment with hexamethonium bromide suppressed the HDR-elicited decrease in ABP, suggesting that the decrease in ABP was induced by the suppression of autonomic neural outflow. The administration of phenoxybenzamine (PB), an alpha-adrenergic antagonist, also eliminated the HDR-elicited decrease in ABP, suggesting that this decrease was elicited by the suppression of alpha-adrenergic vascular tone. To test sympathetic outflow during HDR, renal sympathetic nerve activity (RSNA) and cervical sympathetic trunk (CST) activity (CSTA) were recorded. RSNA was transiently suppressed at 2.3 +/- 0.4 s after HDR onset, followed by a decrease in ABP, suggesting that this decrease was, at least in part, induced by the suppression of sympathetic nerves. CSTA did not change significantly during HDR. These results suggest that HDR suppresses sympathetic nerves in the lower body rather than in the head, which might result in a decrease in ABP. To test the effect of the decrease in ABP due to sympathetic activity on CBF during HDR, changes in CBF during HDR were measured. CBF did not change significantly during HDR in the control group after the administration of an alpha-receptor blocker or after denervation of the CSTs. These results suggest that the impact of the CSTs on CBF is likely to be limited by a rapid increase in CBF due to HDRelicited cephalad fluid shift and that CBF autoregulation proceeds through an alternative mechanism involving the myogenic properties of cerebral vessels.

The immune function of immune organs is extremely crucial for maintaining organism health status, which ultimately affects fish growth. Our previous study has found that dietary supplementation of (2-carboxyethyl) dimethylsulfonium Bromide (Br-DMPT) in non-fish meal (NFM) diet could promote the growth of grass carp (Ctenopharyngodon idella), whereas the underlying reason or mechanism for this results is largely unclear. Herein, we further explored whether dietary supplementation of Br-DMPT promoted fish growth is connected with the enhanced immune function in the immune organs (the head kidney, spleen and skin). In this study, 540 fish (216.49 +/- 0.29 g) were irregularly distributed to six groups with three replicates (30 fish replicate 1) and fed corresponding diets group containing a fish meal (FM) diet group and five different NFM diets supplemented with gradational Br-DMPT (0-520.0 mg/kg level) group for 60 days. After the 60-days feeding trial, 8 fish from each replicate were selected and then conducted a challenge test with A. hydrophila for 14 days. Our results indicated that in the NFM diets, appropriate Br-DMPT: (1) significantly decreased the morbidity of skin haemorrhage and lesion after A. hydrophila infection (P < 0.05). (2) significantly improved the innate and adaptive immune components (lysozyme, complement 3, immunoglobulin M and antibacterial peptides et al.) (P < 0.05). (3) increased the gene expressions of main anti-inflammatory cytokines partially by referring to TOR signalling pathway, and decreased the gene expressions of main pro-inflammatory cytokines partially by referring to NF-kB signalling pathway (P < 0.05). Strikingly, no statistical difference could be found in the most of above immune parameters between 260.0 mg/kg Br-DMPT diet group and FM diet group (P > 0.05). Taken together, in non-fish meal diet, appropriate supplementation of Br-DMPT could improve the disease resistance capacity, non-specific immunity and ameliorate inflammation, and simultaneously could mitigate these adverse effects induced by the non-fish meal diet in fish immune organs. Moreover, this study may be helpful to decipher the underlying mechanisms of how Br-DMPT promote fish growth by immune organs and also provide scientific theoretical evidence for the future application of Br-DMPT as a new immunopotentiator in aquaculture industry.

Thermal polymerization at 210 degrees C provided a PCE/DDSQ framework having a thermal decomposition temperature (516 degrees C) and char yield (70 wt%); these values increased to 600 degrees C and 81 wt%, respectively, after thermal treatment at 420 degrees C. More interestingly, these mesoporous PCE/DDSQ frameworks displayed electrochemical properties better than those of other non-carbonized materials.

Energy Transfer from Perovskite Nanocrystals to Dye Molecules Does Not Occur by FRET

Single formamidinium lead bromide (FAPbBr(3)) perovskite nanocubes, approximately 10 nm in size, have extinction cross sections orders of magnitude larger than single dye molecules and can therefore be used to photoexcite one single dye molecule within their immediate vicinity by means of excitation-energy transfer (EET). The rate of photon emission by the single dye molecule is increased by 2 orders of magnitude under excitation by EET compared to direct excitation at the same laser fluence. Because the dye cannot accommodate biexcitons, NC biexcitons are filtered out by EET, giving rise to up to an order-of-magnitude improvement in the fidelity of photon antibunching. We demonstrate here that, contrary to expectation, energy transfer from the nanocrystal to dye molecules does not depend on the spectral line widths of the donor and acceptor and is therefore not governed by Forster’s theory of resonance energy transfer (FRET). Two different cyanine dye acceptors with substantially different spectral overlaps with the nanocrystal donor show a similar light-harvesting capability. Cooling the sample from room temperature to 5 K reduces the average transition line widths 25-fold but has no apparent effect on the number of molecules emitting, i.e., on the spatial density of single dye molecules being photoexcited by single nanocrystals. Narrow zero-phonon lines are identified for both donor and acceptor, with an energetic separation of over 40 times the line width, implying a complete absence of spectral overlap-even though EET is evident. Both donor and acceptor exhibit spectral fluctuations, but no correlation is apparent between the jitter, which controls spectral overlap, and the overall light harvesting. We conclude that the energy transfer process is fundamentally nonresonant, implying effective energy dissipation in the perovskite donor because of strong electron-phonon coupling of the carriers comprising the exciton. The work highlights the importance of performing cryogenic spectroscopy to reveal the underlying mechanisms of energy transfer in complex donor-acceptor systems.
