Development and Study of Nanoemulsions and Nanoemulsion-Based Hydrogels for the Encapsulation of Lipophilic Compounds

Biocompatible nanoemulsions and nanoemulsion-based hydrogels were formulated for the encapsulation and delivery of vitamin D-3 and curcumin. The aforementioned systems were structurally studied applying dynamic light scattering (DLS), electron paramagnetic resonance (EPR) spectroscopy and viscometry. In vitro studies were conducted using Franz diffusion cells to investigate the release of the bioactive compounds from the nanocarriers. The cytotoxicity of the nanoemulsions was investigated using the thiazolyl blue tetrazolium bromide (MTT) cell proliferation assay and RPMI 2650 nasal epithelial cells as in vitro model. DLS measurements showed that vitamin D-3 and curcumin addition in the dispersed phase of the nanoemulsions caused an increase in the size of the oil droplets from 78.6 +/- 0.2 nm to 83.6 +/- 0.3 nm and from 78.6 +/- 0.2 nm to 165.6 +/- 1.0 nm, respectively. Loaded nanoemulsions, in both cases, were stable for 60 days of storage at 25 degrees C. EPR spectroscopy revealed participation of vitamin D-3 and curcumin in the surfactants monolayer. In vitro release rates of both lipophilic compounds from the nanoemulsions were comparable to the corresponding ones from the nanoemulsion-based hydrogels. The developed o/w nanoemulsions did not exhibit cytotoxic effect up to the concentration threshold of 1 mg/mL in the cell culture medium.

The stability and toxicity of TriCurin microemulsion were tested in an organotypic cervical tissue model. Results TriCurin and its individual compounds inhibit the growth of W12 cells, episomal, type 1 and 2 integrants; the relative order of activity is TriCurin, EGCG, curcumin, or resveratrol. RT-PCR shows that TriCurin activates p53 and suppresses HPV16 mRNAs E1, E2, E4, E6 and E7 at 24 h in W12 cells. Gene expression analysis shows that TriCurin activates pro-apoptotic genes and represses anti-apoptotic genes in HeLa cells. TriCurin in a microemulsion is stable and non-toxic to cervical tissue. The combination of TriCurin and tanshinone IIA exhibits additional synergy against HeLa cells. Conclusions TriCurin, and the combination of TriCurin with tanshinone IIA, are effective against HPV (+) cells. The phytochemical mixture, in the microemulsion-based cream, is a promising therapeutic for the prevention and treatment of cervical cancer.

Effect of vitamin A as a neoadjuvant agent in chemotherapy and photodynamic therapy of Rhabdomyosarcoma cells

Combinational therapy is among the most used treatment modality’s to increase cancer treatment efficacy. It may also reduce side effects, treatment time, and development of drug resistance. The effects of different analogues of vitamin A (VA) as neoadjuvant agent were observed in this study with chemotherapeutic drugs (doxorubicin and methotrexate) and photodynamic therapy (PDT) using 5-ALA and Photogem as photosensitizers in RD cells. The uptake time of photosensitizer was optimized by means of spectrophotometric measurements. Diode laser (lambda = 635 nm +/- 1 nm) was used as an illumination source for PDT. Responses of administered drugs were assessed by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. VA and its analogues exert prophylactic and therapeutic effects. Protective or antagonistic effects (CI > 1) were observed in each of the case. These results propose that the use of VA as a neoadjuvant agent in combinational therapeutic modalities may reduce the efficacy of cancer treatment protocols as well as the existing side effects. Thus, VA is not the successful drug for combinational therapies and under treatment cancer patients should try to avoid its use with oxidative stress induction therapies (e.g. PDT, Radiotherapy, chemotherapy).

A comparative study between in situ screen printed (SPE), carbon paste (CPE) and carbon nanotube modified graphite (CNTME) electrodes was established for the determination of tetra-n-butylammonium bromide (TBAB) in pure and spiked tap water samples. Different experimental conditions as ion-pairing agent type and content, carbon nanotube content, plasticizer, response and soaking time, pH and, temperature effects were studied. The in situ SPEs commingling 6 mg sodium tetraphenylborate (NaTPB, electrode I), 6 mg phosphotungstic acid (PTA, electrode II), 6 mg phosphomolybdic acid (PMA, electrode III), and 12 mg ammonium reineckate (RN, electrode IV) ion-pairing agents displayed the best Nernstian slope values of 60.71 +/- 1.94, 59.75 +/- 0.38, 59.73 +/- 0.76 and 59.90 +/- 0.51 mV decade(-1) over a perceptible spacious concentration range from 1.0 x 10(-5) to 1.0 x 10(-2) mol L-1, respectively. Mitochondrial dysfunction is involved in aristolochic acid I-induced apoptosis in renal proximal tubular epithelial cells

Aristolochic acid (AA) is a compound extracted from the Aristolochia species of herbs. AA exposure is associated with kidney injury known as aristolochic acid nephropathy (AAN). Proximal tubular epithelial cell (PTEC) is the primary target of AA and rich in mitochondria. Recently, increasing evidence suggests that mitochondrial dysfunction plays a critical role in the pathogenesis of kidney disease. However, the status of mitochondrial function in PTEC after exposure to AA remains largely unknown. The aim of this study was to explore the effect of aristolochic acid I (AAI) on cell apoptosis and mitochondrial function in PTEC. Normal rat kidney-52E (NRK-52E) cells were exposed to different concentrations of AAI for different time periods. Cell viability was detected by 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide, cell apoptosis was analyzed by flow cytometry, and the expression of cleaved caspase-3 by Western blotting. Mitochondrial function was evaluated by reactive oxygen species (ROS), mitochondrial membrane potential (MMP), mitochondrial DNA (mtDNA) copy number, and adenosine triphosphate (ATP). It was found that AAI reduced cell viability and increased cell apoptosis in a dose- and time-dependent manner. In parallel to increased apoptosis, NRK-52E cell manifested signs of mitochondrial dysfunction in response to AAI treatment. The data indicated that AAI could increase ROS level, lower MMP, decrease mtDNA copy number, and reduce ATP production. In addition, Szeto-Schiller 31, a mitochondria-targeted antioxidant peptide, attenuated AAI-induced mitochondrial dysfunction and apoptosis. Our study depicted significant aberrant of mitochondrial function in AAI-treated NRK-52E cell, which suggested that mitochondrial dysfunction may be involved in AAI-induced apoptosis in PTEC.

CTAB-Assisted Synthesis of C@Na3V2(PO4)(2)F-3 With Optimized Morphology for Application as Cathode Material for Na-Ion Batteries

Simultaneous Determination of Iron and Cobalt using Spectrophotometry after Catanionic Mixed Micellar Cloud Point Extraction Procedure

A new preconcentration method which utilises a mixture of cationic and anionic surfactants for separation and spectrophotometric determination of iron and cobalt simultaneously has been developed. The metal ions, iron and cobalt were complexed with thiocyante. The hydrophobic complexes of iron and cobalt were then extracted into catanionic mixed micelles of cetyltrimethylammonium bromide (CTAB) and sodium dodecyl sulphate (SDS). Different parameters like concentration of HCl, concentration of thiocyanate, concentrations of the surfactants (CTAB and SDS), equilibration temperature and time were studied to get maximum efficiency. The linear ranges of Fe3+ and Co2+ were found to be 0.139 – 0.838 mu g mL(-1) and 5.89 – 35.4 mu g mL(-1), respectively the detection limits obtained were 1.54 ng mL(-1) and 6.18 ng mL(-1). The developed procedure has been employed for the retrieval of Fe3+ and Co2+ in water samples successfully (tap water and sea water). 98 – 107% recoveries were obtained.
