Analysis of Hop-Derived Thiols in Beer Using On-Fiber Derivatization in Combination with HS-SPME and GC-MS/MS

The quantitation of the hop varietal thiols 4-mercapto-4-methyl-2-pentanone (4MMP), 3-mercapto-1-hexanol (3MH), and 3-mercaptohexylacetate (3MHA) from beer is challenging. This primarily relates to their low concentration (ng/L levels) and their reactivity. Published assays for thiol quantitation from beer include complex and/or time-consuming sample preparation procedures involving manual handling and use reagents that are harmful because they contain mercury. To facilitate thiol analysis from beer, the current article is concerned with the implementation of an automated headspace solid-phase microextraction (HS-SPME) on-fiber derivatization (OFD) approach using 2,3,4,5,6-pentafluorobenzyl bromide followed by gas chromatography-tandem mass spectrometry (GC-MS/MS). Optimization of HS-SPME and MRM conditions was based on a central composite design approach. The final OFD-HS-SPME-GC-MS/MS method yielded limits of quantitation below the sensory thresholds of 4MMP, 3MH, and 3MHA. Method validation and application on beers brewed with German, Australian, and US hops, as well as with added fruits displayed excellent method performance.

Coating of Ca(OH)(2)/gamma-Al2O3 pellets with mesoporous Al2O3 and its application in thermochemical heat storage for CSP plants

Thermochemical heat storage using the Ca(OH)(2) dehydration/hydration reaction for CSP plants requires the development of advanced materials that avoids the breakage shown by conventional CaO pellets. Three strategies of preparation were devised using as coatings alumina gels containing cetyltrimethylammonium bromide as surfactant in neutral (A2 CS) and acid pH (A4 CS) and without surfactant in neutral medium (A3 CS). In both A2 CS and A4 CS materials, denser alumina coatings were attained originating higher crushing strengths, pointing out the positive role played by the surfactant in the gel makeup. SEM images indicated that A2 presented small alumina grains (0.3-3 mm) highly dispersed over their surface, while over A4 CS, long sheets were appreciated. All the materials exhibited high hydration capacities after 10 cycles (>80%, >800 kJ/kg) at 250 Alpha C, but only A2 CS hold its structural integrity that was preserved even after 20 cycles without loss of its hydration capacity. Micro-solvation and counter ion effects on ionic reactions: Activation of potassium fluoride with 18-crown-6 and tert-butanol in aprotic solvents

Ionic reactions have an important solvent effect and the first solvation shell makes the most important contribution for determining the reactivity. In the case of nucleophilic fluorination with potassium fluoride, the solubilization of the salt is another aspect which needs be considered. In the present work, the effect of adding stoichiometric quantity of 18-crown-6 and the bulky alcohol tert-butanol to the reaction of potassium fluoride with alkyl bromide in toluene and acetonitrile solvents was investigated by theoretical methods as molecular dynamics and electronic structure calculations, as well as by experiments. The calculations indicate that the fluoride ion in the IT (18-crown-6) complex interacts with tern-butanol molecules forming aggregates with one (acetonitrite solvent) and three (toluene solvent) tert-butanol molecules. The calculations predict that formation of these aggregates increase the solubilization of KF and produces and important rate acceleration effect in both acetonitrile and toluene solvents, which is experimentally observed. In the concentration around 0.75 mol L-1 of the tert-butanol, the reaction rate is proportional to the alcohol concentration and the rate of alkyl halide decay becomes 10 times higher than the rate without this alcohol. The present results point out that both the interactions of KF with crown ether and tert-butanol molecules have a synergic effect, increasing its reactivity towards S(N)2 reactions. Understanding the Chemistry and Sources of Precipitation Ions in the mid-Brahmaputra Valley of Northeastern India

The chemistry of rainwater over mid-Brahmaputra Valley was studied for three consecutive years (2012-2014; n = 285). The samples were analyzed for major chemical parameters viz. pH, electrical conductivity (EC), and ions (SO42-, NO3-, Cl-, F-, Br-, Ca2+, NH4+, Mg2+, Na+, K+, and Li+), organic acids (HCOO- and CH3COO-) and dissolved organic carbon (DOC). The mean pH for the entire study period was found to be 5.66, which ranged from 4.51 to 7.68, and the volume weighted (VW) mean pH was found to be 5.16. Over 55% of the samples showed pH between 5 and 6, and a few samples had pH 5. Ionic concentration followed the order NH4+ Ca2+ > SO42- > NO3- > Cl- > Na+ > K+ > Mg2+ > H+ > HCO3- > Br- > F- > Li+, indicating dominance of alkaline ions over acidic ions such that 94% of mineral acid was neutralized. The secondary ions, NH4+, SO42-, and NO3-, showed high wet deposition fluxes. The chemistry exhibits explicit seasonality. The airmass clusters of monsoon and non-monsoon seasons, and the associated chemistry varied, which showed influence of long-range transport. The interspecies correlations varied between the monsoon and non-monsoon time samples meaning variation in the source strengths of the contribution sources of the chemical species of the rainwater. Positive Matrix Factorization (PMF) was applied to the data which extracted six factors that explained the sources and chemistry of the rainwater constituents which are of sea, agriculture, coal burning, biomass burning, and secondary origin.

Phospholipase C gamma 2 (PLCG2) has been implicated in the regulation of cell proliferation, transformation, and tumor growth. In this study, we investigate the mechanism of PLCG2 action using a short interference RNA (siRNA) method. The effects of PLCG2 on rat liver BRL-3A cells treated siRNA were studied by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT assay), bromodeoxyuridine (BrdU) labelling assay, flow cytometry method (FCM), quantitative real-time polymerase chain reaction (qRT-PCR) and western blot. The results showed when PLCG2 was reduced, cell vitality and proliferation rate were significantly decreased (p < .05 vs. control). FCM analysis showed that the number of cell division phase (G2 + M) was declined (p < .05 vs. control). RT-PCR and western blot revealed that the expression of signalling related genes NF-kappa B, FOS, JUN and ELK, target genes BCL2, CCNB1 and CCND1 were remarkably down-regulated in cells treated with PLCG2 siRNAs. Based on these results, we conclude PLCG2 plays an important role in rat liver cell proliferation via ERK and NF-kappa B pathway by regulating the expression of BCl2, MYC and CCND1. Monitor application of multi-electrochemical sensor in extracting bromine from seawater

In this paper, a set of online measurement devices of multi-electrochemical sensor was investigated. Combined with industrial distributed control system, it was first applied in extracting bromine from seawater to realize the real-time adjustment of production process parameters. In the process of extracting bromine from seawater, the pH value of acidified raw brine, the addition amount of Cl-2 in the oxidation stage and the addition amount of SO2 in the absorption stage are key parameters to control the whole production process. The multi-electrochemical sensor realized a rapid and high-throughput detection of the above parameters by integrating an all-solid-stage bromide ion selective electrode (Br-ISE), Eh electrode and pH electrode. The Br-ISE and the pH electrode were self-developed electrodes and the Pt electrode was Eh electrode. The pH electrode was used to control the addition amount of H2SO4 during the acidification of the brine. The Eh electrode was used to control the addition amount of Cl-2 during the oxidation stage and the addition amount of SO2 during the absorption stage. The Br-ISE was used to monitor the Br- concentration change in the raw brine. Results showed the optimum range of Eh in the oxidation stage and absorption stage of brine were 950-1000 mV and 580-610 mV, respectively. The application of multi-electrochemical sensor in industrial bromine production can realize real-time control of material addition and save the cost of production.

One-Step Controlled Synthesis of Size-Tunable Toroidal Gold Particles for Biochemical Sensing

Controlling the size and shape of nanoparticles is a major goal in materials science. Here we show the fast, 30 min, controlled one-step synthesis of gold particles (tAUPs) with sizes tunable from 350 nm to 1.7 mu m by using a mixture of surfactant scaffolds made from sodium dodecyl sulfate (SDS) and cetyltrimethylammonium bromide (CTAB). The as-synthesized tAUPs have nanospikes that protrude either inward or outward from the ring cavity. The number of nanospikes can be tuned by a two-step temperature change process to create so-called rough tAUPs. The toroidal gold particle structures exhibit surface plasmon extinction peaks in the near-infrared region and demonstrate a high surface-enhanced Raman scattering (SERS) sensitivity for the detection of 4-mercaptobenzoic acid (4-MBA) molecules. The protruding nanospikes significantly enhance the electromagnetic field oscillating inside the ring cavity. This is confirmed through sensitive detection of 4-MBA molecules as well as by simulation. Rough tAUP structures generate the highest sensitivity with an estimated total enhancement (TE) factor of 3.33 x 10(6), which is 2 orders of magnitude greater than reported by gold nanostars. A variety of different gold structures such as gold nanodendrites, nanowires, and nanochains can be obtained by changing the SDS to CTAB concentration ratios. The unique structures and plasmonic properties of tAUPs hold promise for ultrasensitive biochemical sensing.

Aqueous Two-Phase System Extraction of Polyketide-Based Fungal Pigments Using Ammonium- or Imidazolium-Based Ionic Liquids for Detection Purpose: A Case Study

Demand for microbial colorants is now becoming a competitive research topic for food, cosmetics and pharmaceutics industries. In most applications, the pigments of interest such as polyketide-based red pigments from fungal submerged cultures are extracted by conventional liquid-liquid extraction methods requiring large volumes of various organic solvents and time. To address this question from a different angle, we proposed, here, to investigate the use of three different aqueous two-phase extraction systems using either ammonium- or imidazolium-based ionic liquids. We applied these to four fermentation broths of Talaromyces albobiverticillius (deep red pigment producer), Emericella purpurea (red pigment producer), Paecilomyces marquandii (yellow pigment producer) and Trichoderma harzianum (yellow-brown pigment producer) to investigate their selective extraction abilities towards the detection of polyketide-based pigments. Our findings led us to conclude that (i) these alternative extraction systems using ionic liquids as greener extractant means worked well for this extraction of colored molecules from the fermentation broths of the filamentous fungi investigated here; (ii) tetrabutylammonium bromide, [N4444]Br-, showed the best pigment extraction ability, with a higher putative affinity for azaphilone red pigments; (iii) the back extraction and recovery of the fungal pigments from ionic liquid phases remained the limiting point of the method under our selected conditions for potential industrial applications. Nevertheless, these alternative extraction procedures appeared to be promising ways for the detection of polyketide-based colorants in the submerged cultures of filamentous fungi.
