Background Clinical research has demonstrated that alprostadil has an anti-inflammatory effect; however, to date, its molecular mechanisms remain unclear. This study aimed to examine the anti-inflammatory activity and related mechanisms of alprostadil in lipopolysaccharide (LPS)-treated H9c2 cells. Methods Cell morphology was observed under an inverted light microscope, while cell viability was assessed with the 3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazolium bromide (MTT) assay. Enzyme-linked immunosorbent assays (ELISA) were conducted to study biochemical indicators of cellular damage, such as released lactate dehydrase (LDH) and troponin, and inflammatory cytokine levels including interleukin-1 beta (IL-1 beta), IL-6, IL-17, and tumor necrosis factor-alpha (TNF-alpha). The mRNA expression levels of Wnt5a, c-jun N-terminal kinase (JNK), and nuclear factor kappa B (NF-kappa B) were further investigated by real-time quantitative polymerase chain reaction (RT-PCR). The effects of alprostadil on the Wnt5a/JNK/NF-kappa B pathway in H9c2 cells was examined by Western blotting. Results Alprostadil increased the cell viability of LPS-stimulated H9c2 cells, reduced LDH and troponin production, and attenuated IL-1 beta, IL-6, IL-17, and TNF-alpha secretion. Moreover, alprostadil reduced the mRNA expression of Wnt5a, JNK, and NF-kappa B and decreased the expression of Wnt5a, NF-kappa B, and the ratio of p-JNK/JNK in H9c2 cells treated with LPS. The siWnt5a or JNK inhibitor SP600125 significantly augmented the inhibitory effects of alprostadil on the Wnt5a/JNK/NF-kappa B pathway. Corrosion resistance of alpha- and (alpha plus beta)-Brasses in a descaling solution by a mixture of imidazole and hexadecyltrimethylammonium bromide

Scaling is a major problem in many industrial processes. Oxide scales often formed on the surface of metals and alloys, and it is of high interest to descale it using effective and environment-friendly methods. Herein, the corrosion inhibition effect of the imidazole (IMI) mixed with the Hexadecyltrimethylammonium bromide (CTAB) for alpha- and (alpha + beta)-brasses in a descaling solution (sulfamic (2%) and perchloric (4%) mixture) is studied at 295 K using electrochemical (potentiodynamic polarization measurements (PDP) and electrochemical impedance measurements (EIS)), inductively coupled plasma spectrometry (ICPS) and scanning electron microscopy/energy-dispersive X-ray spectroscopy (SEM/EDX) techniques. Results show that the imidazole/CTAB mixture acts via adsorption at the alloy/solution interface. Potentiodynamic polarization results reveal that the imidazole/CTAB mixture behaves as a mixed type corrosion inhibitor. At the same time, those from EIS tests show that alpha-brass has a higher corrosion resistance compared to that of (alpha + beta)-brass. Under optimum conditions, an inhibition efficiency of 95 % is achieved for both brasses. The morphological characterization by SEM/EDX and ICPS analysis confirms the efficacy of the mixture in protecting alpha- and (alpha + beta)-brasses against corrosion by decreasing the dezincification factor. This study provides a new approach to control the brass loss and achieve desired surface quality using a less corrosive descaling solution and imidazole/CTAB as anti-corrosive additive.

Within each of the four series of complexes characterized by the same thiourea ligand, the Pd(phen) drugs invariably showed the highest anti-proliferative efficacy. This paralleled both a higher intracellular drug accumulation and a more efficient DNA intercalation than all the other metal-bidentate ligand combinations. The consequent inhibition of topoisomerase II activity led to the greatest inhibition of DNA metabolism, evidenced by the inhibition of the expression of the folate cycle enzymes and a marked perturbation of cell-cycle distribution in both cell lines. These findings indicate that the particular interaction of Pd(II) with phenanthroline confers the best pharmacokinetic and pharmacodynamic properties that make this class of DNA intercalators remarkable inhibitors, even of the resistant cell growth.

Ni mesostructured catalysts obtained from rice husk ashes by microwave-assisted synthesis for CO2 methanation

In this work, rice husk ashes were used to obtain a high purity silica precursor to synthesize mesoporous silica frameworks. A novel route for the synthesis of mesocellular silica foam (MCF) was developed using microwave irradiation during the hydrothermal treatment. Cyclohexane and cetyltrimethylammonium bromide were employed as swelling and structure-directing agents, respectively. Ni-based catalysts supported on MCM-41 and MCF mesostructured materials were obtained using two methods, one-pot and incipient wetness impregnation. The catalysts prepared for both methods presented noticeable differences in the metal-support interaction, which were observed by TEM, SAXRD, TPR and XPS. The impregnated catalysts showed preferentially Ni particles inside the pore structure of the support while the one-pot catalysts also showed Ni particles within the silica framework. The catalysts were evaluated in the CO2 methanation reaction between 200 and 500 degrees C at atmospheric pressure. The samples prepared by impregnation showed better catalytic performance than those prepared by the one-pot method. Thereby, the Ni particles highly distributed within the pore improved the CO2 methanation reaction. The Ni/MCF_ch_iwi presented the best catalytic performance reaching CO2 conversions and CH4 selectivities close to the reaction equilibrium, from 350 to 500 degrees C. The Ni/MCF_ch_iwi also showed high structure and catalytic stability, as well as coke resistance formation under reaction conditions.

Development of an ammonium chloride-enhanced thermal-assisted-ESI LC-HRMS method for the characterization of chlorinated paraffins

Simultaneous quantification of short-, medium-, and long-chain chlorinated paraffins (CPs) in environmental matrices is challenging and has received much attention from environmental chemists. In this study, ammonium-chloride-enhanced liquid chromatography coupled with high-resolution mass spectrometry (LC-HRMS) was developed for the first time to quantify CPs in sediments and aqueous samples. Three ionization sources, including atmospheric pressure chemical ionization (APCI), electrospray ionization (ESI), and thermal-assisted-ESI, were employed to examine the performance of ammonium chloride as the chloride ion supply reagent in comparison with traditional chloride ion supply reagent, dichloromethane. Ammonium chloride can be easily used with reversed-phase liquid chromatography (LC), whereas dichloromethane is not compatible with aqueous LC mobile phase. Furthermore, other anion-supply reagents, such as ammonium formate, ammonium acetate, and ammonium bromide, were also tested. It was concluded that the adducts of the CPs with the anions were reversible and could partially dissociate into deprotonated CP ions. The yield of deprotonated CP ions was associated with the gas-phase basicity of the deprotonated CP ions and the corresponding anions. Furthermore, collision-induced dissociation curves were drawn to quantify the stability of anionic CP adducts. The ammonium-chloride-enhanced LC-HRMS was further employed for identifying CPs in sediment samples and coupled with an online SPE method for detecting CPs in aqueous samples. To better understand the process, the effect of DTAB and sodium chloride (NaCl) on interfacial properties of oil/water and oil/water/membrane, such as surface tension, membrane wetting, zeta potential and adhesion map were investigated. It was observed that the surfactant facilitates the oil phase adhesion to the membrane surface, causing a permeate flux reduction. On the other hand, the presence of NaCl in the oily emulsion containing DTAB lead to a permeate flux increase, which indicated that ionic strength modifies the interaction between the oil phase and the membrane surface. Considering the high salinity of the produced water, it is possible to imply that the presence of surfactant can contribute to treatment by ultrafiltration process. Independent of surfactant presence in produced water, the oil and grease (COG) content in the permeate stream was consistently lower than 5 mg/L.
