4.2.1. First-generation strong base process. The mixture was cooled to 40 °C over 2.5 h and water (1.5 L/kg) was added to the solution over 50 min. The resulting slurry was cooled to 25 °C over 1 h and aged for an additional 1 h followed by filtration. The cake was washed with a solution of IPA/water (1:1, 9.17 L) and dried under vacuum (at atmospheric pressure, 50 °C) for 24 h to afford 3 (4.15 kg, 63percent, 12:1 dr).
