Li, Kai;Huang, Shengli;Liu, Tianyu;Jia, Shiqi;Yan, Hailong research published 《 Organocatalytic Asymmetric Dearomatizing Hetero-Diels-Alder Reaction of Nonactivated Arenes》, the research content is summarized as follows. Nonactivated arenes, such as benzene derivatives, are chem. inert due to their intrinsic aromaticity and low polarity. The catalytic asym. dearomatization (CADA, coined by You and co-workers) of the nonactivated arenes represents a formidable challenge. Herein, the authors demonstrate an organocatalytic asym. dearomatizing hetero-Diels-Alder reaction of benzene derivatives The tunable regioselectivity of this strategy allowed delivery of a diversity of stereochem. complex polycyclic compounds, e.g., I, and oxahelicenes, e.g., II, with excellent stereoselectivity. The high complexity and three-dimensionality of the products were crucial for their potential applications in materials science and drug discovery. Mechanistic studies suggested that this reaction proceeded through a chiral tetra-substituted vinylidene ortho-quinone methide (VQM) intermediate, which was extremely active to overcome the loss of aromaticity of benzene derivatives with concomitant chirality transfer.

Li, Jianbin;Huang, Chia-Yu;Han, Jing-Tan;Li, Chao-Jun research published 《 Development of a quinolinium/cobaloxime dual photocatalytic system for oxidative C-C cross-couplings via H2 release》, the research content is summarized as follows. Designing mol. photocatalysts for potent photochem. reactivities ranks among the most challenging but rewarding endeavors in synthetic photochem. Herein, we document a quinoline-based organo-photoredox catalyst, 2,4-bis(4-methoxyphenyl)quinoline (DPQN2,4-di-OMe), that could be assembled via the facile aldehyde-alkyne-amine (A3) couplings. Unlike the reported photocatalysts, which impart their photoreactivities as covalently linked entities, our mechanistic studies suggested a distinct proton activation mode of DPQN2,4-di-OMe. Simply upon protonation, DPQN2,4-di-OMe could reach a highly oxidizing excited state under visible-light irradiation (E*1/2 = +1.96 V vs a standard calomel electrode, SCE). On this basis, the synergistic merger of DPQN2,4-di-OMe and cobaloxime formulated an oxidative cross-coupling platform, enabling the Minisci alkylation and various C-C bond-forming reactions with a diverse pool of radical precursors in the absence of chem. oxidants. The catalytic loading of DPQN2,4-di-OMe could be minimized to 0.025 mol % (TON = 3360), and a polymer-supported photocatalyst, DPQN2,4-di-OR@PS, was prepared to facilitate catalyst recycling (at a 0.50 mmol % loading and up to five times without significant loss of photosynthetic efficiency).

The sphingosine 1-phosphate (S1P) signaling pathway is an attractive target for pharmacol. manipulation due to its involvement in cancer progression and immune cell chemotaxis. The synthesis of S1P is catalyzed by the action of sphingosine kinase 1 or 2 (SphK1 or SphK2) on sphingosine and ATP. While potent and selective inhibitors of SphK1 or SphK2 have been reported, development of potent dual SphK1/SphK2 inhibitors are still needed. Towards this end, we report the structure-activity relationship profiling of 2-(hydroxymethyl)pyrrolidine-based inhibitors with 22d being the most potent dual SphK1/SphK2 inhibitor (SphK1 Ki = 0.679μM, SphK2 Ki = 0.951μM) reported in this series. 22D inhibited the growth of engineered Saccharomyces cerevisiae and decreased S1P levels in histiocytic lymphoma myeloid cell line (U937 cells), demonstrating inhibition of SphK1 and 2 in vitro. Mol. modeling studies of 22d docked inside the Sph binding pocket of both SphK1 and SphK2 indicate essential hydrogen bond between the 2-(hydroxymethyl)pyrrolidine head to interact with aspartic acid and serine residues near the ATP binding pocket, which provide the basis for dual inhibition. In addition, the dodecyl tail adopts a “J-shape” conformation found in crystal structure of sphingosine bound to SphK1. Collectively, these studies provide insight into the intermol. interactions in the SphK1 and 2 active sites to achieve maximal dual inhibitory activity.

Li, Gen;Miller, Steven P.;Radosevich, Alexander T. research published 《 PIII/PV=O-Catalyzed Intermolecular N-N Bond Formation: Cross-Selective Reductive Coupling of Nitroarenes and Anilines》, the research content is summarized as follows. An organophosphorus-catalyzed method for the synthesis of unsym. hydrazines e.g., I by cross-selective intermol. N-N reductive coupling is reported. This method employs a small ring phosphacycle (phosphetane) catalyst II together with hydrosilane as the terminal reductant to drive reductive coupling of nitroarenes e.g., 1-methanesulfonyl-4-nitrobenzene and anilines e.g., 2,3,4,5-tetrahydro-1H-1-benzazepine with good chemoselectivity and functional group tolerance. 4-Bromophenylmethylamine is an organic compound that has anti-inflammatory properties and is used as a pharmaceutical. It belongs to the group of amines. The hydrolysis of 4-bromophenylmethylamine by hydrochloric acid produces phenol and bromamine (NHBr). The reaction system can be used to synthesize a number of compounds, including anilines, benzofurans, and other aromatic compounds. 4-Bromophenylmethylamine reacts with muscle tissue in a similar manner as acetaminophen does. This drug also has been shown to have significant effects on the energy metabolism in the muscles of rats that are given carbon source., 6911-87-1.

Organic compounds having carbon bonded to bromine are called organic bromides. 6911-87-1, formula is C7H8BrN, Name is 4-Bromo-N-methylaniline. Depending on the type of carbon to which the bromine is bonded, organic bromide could be alkyl, alkenyl, alkynyl, or aryl. Category: bromides-buliding-blocks.

Li, Gen;Qin, Ziyang;Radosevich, Alexander T. research published 《 P(III)/P(V)-Catalyzed Methylamination of Arylboronic Acids and Esters: Reductive C-N Coupling with Nitromethane as a Methylamine Surrogate》, the research content is summarized as follows. The direct reductive N-arylation of nitromethane by organophosphorus-catalyzed reductive C-N coupling with arylboronic acid derivatives is reported. This method operates by the action of a small ring organophosphorus-based catalyst (1,2,2,3,4,4-hexamethylphosphetane P-oxide) together with a mild terminal reductant hydrosilane to drive the selective installation of the methylamino group to (hetero)aromatic boronic acids and esters. This method also provides for a unified synthetic approach to isotopically labeled N-methylanilines from various stable isotopologues of nitromethane (i.e., CD3NO2, CH315NO2, and 13CH3NO2), revealing this easy-to-handle compound as a versatile precursor for the direct installation of the methylamino group.

Li, Gen team published research in Journal of the American Chemical Society in 2020 | 244205-40-1

244205-40-1, 2-Bromophenylboronic Acid is used as an inhibitor of the hormone sensitive lipase.
2-Bromophenylboronic acid, also known as 2-Bromophenylboronic acid, is a useful research compound. Its molecular formula is C6H6BBrO2 and its molecular weight is 200.83 g/mol. The purity is usually 95%.
2-Bromophenylboronic acid is a glucose monitoring agent that has a ruthenium complex with an acidic environment. The nitro group and the amines are in close proximity to the boron center, and this proximity leads to a high nucleophilic character of the molecule. This reactivity allows 2-bromophenylboronic acid to be used as a fluorescence probe for acidic environments. 2-Bromophenylboronic acid also inhibits secretase enzymes, which are involved in Alzheimer’s disease and other neurodegenerative disorders. It is an inhibitor of γ-secretase, which is responsible for cleaving the amyloid precursor protein (APP), and it has shown efficacy against biphenyl, an anticancer drug that binds to benzodiazepine receptors. 2-Bromophenylboronic acid is also an enantiopure compound because all four substituents are different from each other., Computed Properties of 244205-40-1
