A variety of different stilbene derivatives were synthesized with exclusive formation of the (E)-isomers and isolated yields up to 97%. However, the influence of steric alkyl chain length has not been studied for the diastereoselectivity. In this work, a new strategy for the racemic synthesis of β-Me alcs. R1CHMeCH(OH)R2 (R1 = Et, n-Pr, n-pentyl; R2 = Et, n-Pr, n-Bu, n-pentyl, n-hexyl, n-heptyl, n-octyl) to obtain various diastereomer ratios using the Felkin-Anh model is reported. The addition of alkyl Grignard reagents R2MgBr to α-Me aldehydes R1CHMeCHO afforded the products with diastereomer ratios of threo/erythro ≈ 2:1, while the reduction in structurally related ketones R1CHMeC(O)R2 using LiAlH4 afforded ratios of threo/erythro ≈ 1:1. The exptl. data showed no effect of alkyl chain length on either side on the stereoselectivity of adducts. All synthesized analogs were evaluated for attractiveness to Rhynchophorus ferrugineus weevils in the field. Five novel derivatives, including two alcs. and three ketones, were found to attract weevils in the field trials. Synthesis of tubulysin analogs, containing an N-Me substituent on tubuvaline-amide together with the replacement of either the hydrophobic N-terminal N-Me pipecolic acid (Mep) or at both N- and C- terminal peptides with available heteroaromatic acids and an unsaturated tubuphenylalanine moiety, resp., were described. The in vitro cytotoxic activity by SRB assay on five cancer cell lines for sixteen tubulysins was evaluated.
