Lawer, Aggie;Rossi-Ashton, James A.;Stephens, Thomas C.;Challis, Bradley J.;Epton, Ryan G.;Lynam, Jason M.;Unsworth, William P. research published 《 Internal Nucleophilic Catalyst Mediated Cyclisation/Ring Expansion Cascades for the Synthesis of Medium-Sized Lactones and Lactams》, the research content is summarized as follows. A strategy for the synthesis of medium-sized lactones and lactams from linear precursors is described in which an amine acts as an internal nucleophilic catalyst to facilitate a novel cyclisation/ring expansion cascade sequence. This method obviates the need for the high-dilution conditions usually associated with medium-ring cyclisation protocols, as the reactions operate exclusively via kinetically favorable “normal”-sized cyclic transition states. This same feature also enables biaryl-containing medium-sized rings to be prepared with complete atroposelectivity by point-to-axial chirality transfer.

Lavinda, Olga;Witt, Collin H.;Woerpel, K. A. research published 《 Origin of High Diastereoselectivity in Reactions of Seven-Membered-Ring Enolates》, the research content is summarized as follows. Unlike many reactions of their six-membered-ring counterparts, the reactions of chiral seven-membered-ring enolates e.g., 5-(tert-Butyl)-1-methylazepan-1-one are highly diastereoselective. Diastereoselectivity was observed for a range of substrates, including lactam, lactone, and cyclic ketone derivatives The stereoselectivity arises from torsional and steric interactions that develop when electrophiles approach the diastereotopic π-faces of the enolates, which are distinguished by subtle differences in the orientation of nearby atoms of the ring.

The Trypanosoma cruzi (T. cruzi) parasite is the cause of Chagas disease, a neglected disease endemic in South America. The life cycle of the T. cruzi parasite is complex and includes transitions between distinct life stages. This change in phenotype (without a change in genotype) could be controlled by epigenetic regulation, and might involve the bromodomain-containing factors 1-5 (TcBDF1-5). However, little is known about the function of the TcBDF1-5. Here we describe a fragment-based approach to identify ligands for T. cruzi bromodomain-containing factor 3 (TcBDF3). We expressed a soluble construct of TcBDF3 in E. coli, and used this to develop a range of biophys. assays for this protein. Fragment screening identified 12 compounds that bind to the TcBDF3 bromodomain. On the basis of this screen, we developed functional ligands containing a fluorescence or 19F reporter group, and a photo-crosslinking probe for TcBDF3. These tool compounds will be invaluable in future studies on the function of TcBDF3 and will provide insight into the biol. of T. cruzi.

Despite the widespread application of hypervalent iodines, the corresponding λ3-bromanes are less explored. Herein the authors report a general, safe, and high-yielding strategy to access cyclic diaryl λ3-bromanes. These unique compounds feature reactivity that is appealing and complementary to that of λ3-iodanes, generating arynes under mild reaction conditions and in the presence of a weak base. Accordingly, formal meta-selective transition-metal-free C-O and C-N couplings may be achieved. Mechanistic studies unambiguously support the aryne generation mechanism.

Optimization of selectivity against related enzymes as well as enhancement of mean residence time of the adduct were critical to the identification of compounds with potent cellular pathway inhibition and ultimately a prolonged pharmacodynamic effect and efficacy in preclin. tumor models, culminating in the identification of the clin. mol. The nanomaterial exhibited an excellent catalytic activity in the synthesis of selenoesters of amino acids through the reaction of corresponding amino acyl chloride derivatives with the in situ-generated NaHSe, followed by addition of α-bromo esters. Wide ranges of unsym. selenoester derivatives of amino acids were afforded with good to excellent yields, and the structures were characterized by high-resolution mass spectroscopy, 1H and 13C NMR spectroscopy, and Fourier transform IR spectroscopy techniques. Furthermore, nano-CeO2 exhibits tremendous catalytic activity in the production of biodiesel from Annona squamosa oil and achieved around 89% biodiesel yield. The biodiesel produced satisfies ASTM standards

Reference of 5445-17-0, Methyl 2-bromopropionate, also known as Methyl 2-bromopropionate, is a useful research compound. Its molecular formula is C4H7BrO2 and its molecular weight is 167 g/mol. The purity is usually 95%.
Methyl 2-bromopropionate is used in the synthesis of poly(ADP-Ribose)polymerase inhibitors derived from benzoxazin-3-one. Also used in the synthesis of 5-HT2C antagonists affecting serotonin levels.
Methyl 2-bromopropanoate is a chemical compound that can be synthesized in an asymmetric manner. The reaction of methyl 2-bromopropanoate with hydrochloric acid gives the corresponding carboxylic acid, methyl propanoate, and hydrogen bromide in a 1:1 ratio. It has been shown that methyl 2-bromopropanoate is a potential catalyst for the reduction of chloride to chloride ion via the borohydride reduction method. Methyl 2-bromopropanoate has also been used as a model system for studying halides and copper complexes. Magnetic resonance spectroscopy studies have revealed that this chemical compound has a high redox potential and kinetic properties., 5445-17-0.

Lacivita, Enza;Niso, Mauro;Mastromarino, Margherita;Garcia Silva, Andrea;Resch, Cibell;Zeug, Andre;Loza, Maria I.;Castro, Marian;Ponimaskin, Evgeni;Leopoldo, Marcello research published 《 Knowledge-Based Design of Long-Chain Arylpiperazine Derivatives Targeting Multiple Serotonin Receptors as Potential Candidates for Treatment of Autism Spectrum Disorder》, the research content is summarized as follows. Autism spectrum disorder (ASD) includes a group of neurodevelopmental disorders characterized by core symptoms such as impaired social interaction and communication, repetitive and stereotyped behaviors, and restricted interests. To date, there are no effective treatments for these core symptoms. Several studies have shown that the brain serotonin (5-HT) neurotransmission system is altered in both ASD patients and animal models of the disease. Multiple pieces of evidence suggest that targeting 5-HT receptors may treat the core symptoms of ASD and associated intellectual disabilities. In fact, stimulation of the 5-HT1A receptor reduces repetitive and restricted behaviors; blockade of the 5-HT2A receptor reduces both learning deficits and repetitive behavior, and activation of the 5-HT7 receptor improves cognitive performances and reduces repetitive behavior. On such a basis, we have designed novel arylpiperazine derivatives pursuing unprecedently reported activity profiles: dual 5-HT7/5-HT1A receptor agonist properties and mixed 5-HT7 agonist/5-HT1A agonist/5-HT2A antagonist properties. Seventeen new compounds were synthesized and tested in radioligand binding assay at the target receptors. We have identified the dual 5-HT1AR/5-HT7R agonists 8c and 29 and the mixed 5-HT1AR agonist/5-HT7R agonist/5-HT2AR antagonist 20b. These compounds are metabolically stable in vitro and have suitable central nervous system druglike properties.
