Zhang, Zeshuai;Wen, Fei;Liu, Haiyan;Li, Zheng research published 《 Selective N-Monovinylation of Primary Aromatic Amides Using Calcium Carbide as an Alkyne Source》, the research content is summarized as follows. An efficient method for the selective N-monovinylation of primary aromatic amides using calcium carbide as an alkyne source was described. The salient features for this protocol were the use of inexpensive, easy-to-handle solid alkyne source, high chemoselectivity, transition metal catalyst-free, good functional group tolerance, and simple work-up procedures. These reactions was also be extended to the gram-scale level.

Tetra-substituted alkenyl nitriles, e.g. I, are obtained providing useful boron-dienenitrile building blocks that can be further functionalized. The utility of our method has been demonstrated with the synthesis of Satigrel, an anti-platelet aggregating agent.

Zhang, Yi;Li, Chenglin;Zhu, Qiuwen;Liang, Renjie;Xie, Chang;Zhang, Shufang;Hong, Yi;Ouyang, Hongwei research published 《 A long-term retaining molecular coating for corneal regeneration》, the research content is summarized as follows. Corneal injuries will cause corneal surface diseases that may lead to blindness in millions of people worldwide. There is a tremendous need for biomaterials that can promote corneal regeneration with practical feasibility. Here we demonstrate a strategy of a protein coating for corneal injury regeneration. We synthesize an o-nitrosobenzaldehyde group (NB)-modified gelatin (GelNB), which could adhere directly to the corneal surface with covalent bonding to form a thin mol. coating. The mol. coating could avoid rapid clearance and provide a favorable environment for cell migration, thereby effectively accelerating corneal repair and regeneration. The histol. structure of the regenerated cornea is more similar to the native cornea. This mol. coating can be used conveniently as an eye drop solution, which makes it a promising strategy for corneal regeneration.

Zhang, Yao;Ma, Jiawei;Chen, Jian;Meng, Lingpu;Liang, Yong;Zhu, Shaolin research published 《 A relay catalysis strategy for enantioselective nickel-catalyzed migratory hydroarylation forming chiral α-aryl alkylboronates》, the research content is summarized as follows. Ligand-controlled reactivity plays an important role in transition-metal catalysis, enabling a vast number of efficient transformations to be discovered and developed. However, a single ligand is generally used to promote all steps of the catalytic cycle (e.g., oxidative addition, reductive elimination), a requirement that makes ligand design challenging and limits its generality, especially in relay asym. transformations. Herein it is hypothesized that multiple ligands with a metal center might be used to sequentially promote multiple catalytic steps, thereby combining complementary catalytic reactivities through a simple combination of simple ligands. With this relay catalysis strategy (L/L*), the first highly regio- and enantioselective remote hydroarylation process is reported. By synergistic combination of a known chain-walking ligand and a simple asym. cross-coupling ligand with the nickel catalyst, enantioenriched α-aryl alkylboronates could be rapidly obtained as versatile synthetic intermediates through this formal asym. remote C(sp3)-H arylation process.

Targeting BCL-XL via PROTACs is a promising strategy in reducing BCL-XL inhibition associated platelet toxicity. Recently, we reported potent BCL-XL PROTAC degraders that recruit VHL or CRBN E3 ligase. However, low protein expression or mutation of the responsible E3 ligase has been known to result in decreased protein degradation efficiency of the corresponding PROTACs. To overcome these mechanisms of resistance, PROTACs based on recruiting alternative E3 ligases could be generated. Thus, we designed and synthesized a series of PROTACs that recruit IAP E3 ligases for BCL-XL degradation Among those PROTACs, compound 8a(I) efficiently degrades BCL-XL in malignant T-cell lymphoma cell line MyLa 1929 while CRBN-based PROTACs that have high potency in other cancer cell lines show compromised potency, likely due to the low CRBN expression. Moreover, compared with the parent compound ABT-263, PROTAC 8a shows comparable cell killing effects in MyLa 1929 cells whereas the on-target platelet toxicity is significantly reduced. Our findings expand the anti-tumor spectra of BCL-XL degraders and further highlight the importance of selecting suitable E3 members to achieve effective cellular activity.

Product Details of C5H9BrO2, Methyl 4-bromobutyrate,also as known as 4-Bromobutyric acid methyl ester, is a useful research compound. Its molecular formula is C5H9BrO2 and its molecular weight is 181.03 g/mol. The purity is usually 95%.
4-Bromobutyric acid methyl ester is a synthetic compound that can be used to inhibit the activity of the G1 phase cyclin-dependent kinases. It has been shown to inhibit protein synthesis by alkylating the amino groups of proteins and fatty acids. 4-Bromobutyric acid methyl ester also inhibits the growth of cancer cell lines, such as renal carcinoma cells. The mechanism of action for this drug is not well understood, but it may be due to its ability to bind with monoclonal antibodies and enter kidney cells by passive diffusion., 4897-84-1.

Organobromine compounds, also called organobromides, are organic compounds that contain carbon bonded to bromine. 4897-84-1, formula is C5H9BrO2, The most pervasive is the naturally produced bromomethane. Product Details of C5H9BrO2

Zhang, Xu;Fang, Wan-Yin;Lekkala, Ravindar;Tang, Wenjian;Qin, Hua-Li research published 《 An Easy, General and Practical Method for the Construction of Alkyl Sulfonyl Fluorides》, the research content is summarized as follows. A visible light-induced reductive addition of 1°, 2° and 3° alkyl iodides to ethenesulfonyl fluoride, employing Hantzsch ester as a hydrogen source at room temperature was developed for the synthesis of alkyl sulfonyl fluorides RCH2CH2SO2F [R = cyclohexyl, Bn, 1-adamantyl, etc.]. This method featured a wide substrate scope and great functional group compatibility, providing facile and robust process to alkyl sulfonyl fluorides including enzyme inhibitors, natural products and drugs derivatives in up to 99% yield. Further derivatization of resultant aliphatic sulfonyl fluorides was also achieved through sulfur fluoride exchange (SuFEx) reactions to deliver sulfonates and sulfonamides as privileged motifs for medicinal chem.
