Herein, a novel series of dual histone deacetylase (HDAC) and vascular endothelial growth factor receptor (VEGFR) inhibitors were designed, synthesized and biol. evaluated based on previously reported pazopanib-based HDAC and VEGFR dual inhibitors. Most target compounds showed significant HDAC1, HDAC6 and VEGFR2 inhibition, which contributed to their potent antiproliferative activities against multiple cancer cell lines and significant antiangiogenic potencies in both human umbilical vein endothelial cell (HUVEC) tube formation assays and rat thoracic aorta ring assays. Further HDAC selectivity evaluations indicated that hydroxamic acids 5 and 9e possessed HDAC isoform selectivity profiles similar to that of the approved HDAC inhibitor suberoylanilide hydroxamic acid(SAHA), while hydrazide12 presented an HDAC isoform selectivity profilesimilar to that of the clin. HDAC inhibitor MS-275. The VEGFR inhibition profiles of 5, 9e and 12 were similar to that of the approved VEGFR inhibitor pazopanib. The intracellular target engagements of Compounds 5 and 12 were confirmed by western blot anal. The metabolic stabilities of 5, 9e and 12 in mouse liver microsomes were inferior to that of pazopanib. These dual HDAC and VEGFR inhibitors provide lead compounds for further structural optimization to obtainpolypharmacol. anticancer agents.

Xuan, Jun;Haelsig, Karl T.;Sheremet, Michael;Machicao, Paulo A.;Maimone, Thomas J. research published 《 Evolution of a Synthetic Strategy for Complex Polypyrrole Alkaloids: Total Syntheses of Curvulamine and Curindolizine》, the research content is summarized as follows. Structurally unprecedented antibacterial alkaloids containing multiple electron-rich pyrrole units have recently been isolated from Curvularia sp. and Bipolaris maydis fungi. This article documents the evolution of a synthetic program aimed at accessing the flagship metabolites curvulamine and curindolizine, which are presumably a dimer and trimer of a C10N biosynthetic building block, resp. Starting with curvulamine, we detail several strategies to merge two simple, bioinspired fragments, which while ultimately unsuccessful, led us toward a pyrroloazepinone building block-based strategy and an improved synthesis of this 10π-aromatic heterocycle. A two-step annulation process was then designed to forge a conserved tetracyclic bis-pyrrole architecture and advanced into a variety of late-stage intermediates; unfortunately, however, a failed decarboxylation thwarted the total synthesis of curvulamine. By tailoring our annulation precursors, success was ultimately found through the use of a cyanohydrin nucleophile which enabled a 10-step total synthesis of curvulamine I. Attempts were then made to realize a biomimetic coupling of curvulamine with an addnl. C10N fragment to arrive at curindolizine, the most complex family member. Although unproductive, we developed a 14-step total synthesis of this alkaloid, II, through an abiotic coupling approach. Throughout this work, effort was made to harness and exploit the innate reactivity of the pyrrole nucleus, an objective which has uncovered many interesting findings in the chem. of this reactive heterocycle.

Nitric oxide (NO) has an important class of endogenous diat. mols. that play a key regulatory role in many physiol. and biochem. processes. However, the type of nitrosamine NO donor stimulated by light has many advantages compared to the conventional NO donors such as diazeniumdiolates and S-nitrosothiols compounds, including easy synthesis, good stability, and controllable release. In addition, NO release can be regulated by light induction with a built-in calibration mechanism fluorescence. Here, we report that the migration and proliferation of human umbilical vein vascular endothelial cells could be accelerated by the light-triggered NO donors, leading to the angiogenesis. Meanwhile, the screened NO donor 3a with Levofloxacin (Lev) showed synergistic effects to eradicate Methicillin-resistant Staphylococcus aureus (MRSA) biofilms in vitro and treat bacteria-infected wound in vivo.

The sensor showed a high selectivity for Al3+ and Cu2+ in methanol solution at pH = 5 and was not disturbed by other competing metal ions. Furthermore, based on the equation 3σ/slope, the detection limits of sensor for Al3+ and Cu2+ were calculated to be 3.1 x 10-10 M and 9.8 x 10-11 M, resp. Addnl., the association constants of X[Al3+] and X[Cu2+] were also determined to be 2.3 x 104 M-1 and 6.1 x 104 M-1 on basis of Benesi-Hildebrand equation. In addition, the titration experiment of fluorescence and mass spectrometry showed that sensor was combined with Al3+ or Cu2+ both in 1:1 ratio. Moreover, the optimized structure and energy calculations of X, X[Al3+] and X[Cu2+] were obtained by Gaussian software based on the basis set of B3LYP/6-31 G(d) and B3LYP/LANL2DZ. And, the sensor successfully detected Al3+ and Cu2+ in the real water sample with a satisfactory recovery (91.4 % – 107.1 %) and RSD values (0.66 % – 1.91 %).

Two types of “parental” BN-dibenzo[f,k]tetraphenes (BNDBT-1 and BNDBT-2) have been synthesized via a transition-metal-catalyzed tandem cross-coupling reaction as key steps. Both BNDBT-1 and BNDBT-2 are fully characterized; one of them is unambiguously confirmed by a single X-ray crystal structure. Compared to its all-carbon analog DBT, BNDBT-1 and BNDBT-2 exhibit a higher HOMO (HOMO) and lower LUMO (LUMO) energy, while the BN doping position slightly influences the HOMO and LUMO energies of BNDBT-1 and BNDBT-2. Both BNDBT-1 and BNDBT-2 exhibit red-shifted absorption and emission spectra and higher emission efficiencies, as compared to their carbonaceous analog DBT. Moreover, organic light emitting diodes were fabricated using BNDBT-1 and BNDBT-2 as emitters, demonstrating their potential applications.

Compounds 8c (I, IC50 = 0.05μM, MCF-7; 0.09μM, NCI-H157) and 8h (II, IC50 = 0.01μM, MCF-7; 0.02μM, NCI-H157) exhibited certain antitumor activities comparable to those of Taxol. The urea analogs of pretubulysin might represent a promising scaffold for the further development of novel antitumor drugs.

Synthetic Route of 70-23-5, Ethyl bromopyruvate molecular formula is C5H7BrO3 and its molecular weight is 195.01 g/mol. The purity is usually 95%.

Ethyl bromopyruvate is used in a synthesis of thioxothiazolidines from carbon disulfide and primary amines.

Ethyl bromopyruvate is a chemical inhibitor that inhibits the enzyme pyruvate dehydrogenase, which is responsible for the conversion of pyruvic acid to acetyl-CoA. This inhibition leads to a decrease in ATP levels and can cause metabolic disorders. Ethyl bromopyruvate is used as an anthelmintic drug and in asymmetric synthesis. It is also used in the synthesis of thiostrepton, an antibiotic that has been shown to have antimicrobial activity against Gram-positive bacteria, including Staphylococcus aureus and Streptococcus pneumoniae., 70-23-5.

402-49-3, 4-Trifluoromethylbenzyl bromide is a useful research compound. Its molecular formula is C8H6BrF3 and its molecular weight is 239.03 g/mol. The purity is usually 95%.
4-Trifluoromethylbenzyl bromide is a choline derivative that acts as an anticancer agent. It is structurally similar to the anticancer drug doxorubicin, which has been shown to be effective against breast cancer and leukemia. 4-Trifluoromethylbenzyl bromide interacts with cellular proteins, including choline kinase, and inhibits the mitochondrial pathway. This leads to cell death through apoptosis. The molecule also interacts with nucleotide bases such as thymine and cytosine in DNA, inhibiting transcription and replication. 4-Trifluoromethylbenzyl bromide binds strongly to the hydroxyl group of cholesterol by an electrophilic substitution mechanism to form a covalent bond with its hydroxy group. The molecule can also bind to chloride ions by an ionic bond., Reference of 402-49-3

Xu, Xi;Chang, Xiujin;Huang, Jingxuan;Zhang, Di;Wang, Min;Jing, Tian;Zhuang, Yu;Kou, Junping;Qiu, Zhixia;Wang, Jubo;Li, Zhiyu;Bian, Jinlei research published 《 Discovery of novel glutaminase 1 allosteric inhibitor with 4-piperidinamine linker and aromatic heterocycles》, the research content is summarized as follows. Glutaminase 1 (GLS1) is overexpressed in multiple types of malignant tumors and is viewed as a promising target in cancer therapy. Thus, the discovery for small-mol. GLS1 inhibitors is urgent. Based on the authors’ previous study of compound I, a potent GLS1 allosteric inhibitor yet with a limited metabolic stability, a structure-based optimization was carried out, with a series of novel GLS1 allosteric inhibitors rationally designed, synthesized and biol. evaluated. Such endeavor has culminated in the identification of compound II, a promising GLS1 allosteric inhibitor with a 4-piperidinamine linker and aromatic heterocycles. Compound II displayed robust GLS1 binding affinity, superior liver microsome metabolic stability, and moderate anti-tumor activity (TGI: 47.5%) in HCT116 xenograft model with no considerable toxicity in vivo. The mechanism underlying the anti-proliferative effect of compound II on HCT116 cells was studied, revealing that the compound blocked the glutamine metabolism, induced the production of ROS, and triggered apoptosis. These findings merit further investigation of compound II as a targeted cancer therapeutic.
