Tunable electrochemical synthesis of pyrrole-based adsorbents

Pyrrole-based adsorbents were developed electrochemically using systems: (i) pyrrole-potassium bromide, (ii) pyrrole-KBr-an anionic surfactant sodium dodecyl sulfate, and (iii) pyrrole-KBr-NaDS-Prussian blue. Growth kinetics and morphological studies were carried out at different experimental conditions. The study revealed the formation of nanosized particles, suitable for the removal of methylene blue as evident by high adsorption capacity = 447.7 mg g(-1). The effect of adsorption dosage, contact time, pH, and temperature on methylene blue removal has been studied. Preparation and characterisation of magnetosomes based drug conjugates for cancer therapy

The authors report a novel, effective and enhanced method of conjugating anticancer drug, paclitaxel and gallic acid with magnetosomes. Here, anticancer drugs were functionalised with magnetosomes membrane by direct and indirect (via crosslinkers: glutaraldehyde and 3-aminopropyltriethoxysilane) adsorption methods. The prepared magnetosome-drug conjugates were characterised by Fourier transform infrared, zeta potential, field-emission scanning electron microscope and thermogravimetric analysis/differential scanning calorimetry. The drug-loading efficiency and capacity were found to be 87.874% for paclitaxel (MP) and 71.3% for gallic acid (MG), respectively as calculated by ultraviolet spectroscopy and high-performance liquid chromatography. The drug release demonstrated by the diffusion method in phosphate buffer (PBS), showing a prolonged drug release for MP and MG, respectively. The cytotoxicity effect of the MP and MG displayed cytotoxicity of 69.71%, 55.194% against HeLa and MCF-7 cell lines, respectively. The reactive oxygen species, acridine orange and ethidium bromide and 4, 6-diamidino-2-phenylindole staining of the drug conjugates revealed the apoptotic effect of MP and MG. Further, the regulation of tumour suppressor protein, p53 was determined by western blotting which showed an upregulation of p53. Comparatively, the magnetosome-drug conjugates prepared by direct adsorption achieved the best effects on the drug-loading efficiency and the increased percentage of cancer cell mortality and the upregulation of P53. The proposed research ascertains that magnetosomes could be used as effective nanocarriers in cancer therapy.

The thermal/pH-sensitive drug delivery system encapsulated by PAA based on hollow hybrid nanospheres with two silicon source

The synthesis of drug delivery systems based on hollow mesoporous silica nanoparticles (MSNs) is still a major challenge. In this work, the hollow hybrid MSNs were successfully prepared by cetyltrimethylammonium bromide-directed sol-gel process and one-step hydrothermal treatment process. The hollow hybrid MSNs had hybrid ethane-bridged frameworks with the uniform particle size (250 nm) and mesoporous pore diameter (3.7 nm). The polyacrylic acid (PAA) encapsulated drug delivery system based on hollow hybrid MSNs was prepared by using silanization, surface modification, doxorubicin hydrochloride (DOX) loading, and PAA coating. Drug encapsulation and release behavior at different temperatures and pH were studied by using DOX as a model guest molecule. The results displayed that the modified hollow ethane-bridged MSNs possessed good biocompatibility and excellent thermal/pH-dual-sensitive drug release property. This novel thermal/pH-sensitive drug delivery system based on hollow ethane-bridged MSNs has the advantages of feasible synthesis, no cytotoxicity, and good drug loading capacity, which may have potential applications in the anticancer therapy.

Fabrication of Cationic Surfactant (Conventional/Gemini) Functionalized Gr@ZrO2 Nanocomposite with Faster Adsorbability of an Anionic Azo Dye from Aqueous Solution

Graphene zirconium oxide nanocomposite (Gr@ZrO2-NC) has been functionalized with cetyltrimethylammonium bromide (CTAB or CT) or butanediyl-1,4, bis (N, N-hexadecyl ammonium) dibromide (cationic gemini surfactant (CGS)). Structural, morphological, optical and thermal properties of synthesized adsorbents (Gr@ZrO2-NC, CT-Gr@ZrO2-NC and CGS-Gr@ZrO2-NC) are investigated using XRD, FTIR, electron microscopy and TGA. CGS-Gr@ZrO2-NC showed faster removal efficacy (90 % within the first 25 min) for anionic azo dyes (methyl orange, MO and Congo red, CR) compared to other adsorbents. This effect may be due to the creation of stronger hydrophobic region at the surface of adsorbent with CGS. Effects of concentration, contact time and pH were also seen. The adsorption efficacy remained better even in the simultaneous presence of MO and CR. Adsorption data with time showed that the pseudo-second-order kinetic model has been obeyed. Adsorbents were regenerated successfully with ethylene glycol (similar to 83 %).

Analysis of Hop-Derived Thiols in Beer Using On-Fiber Derivatization in Combination with HS-SPME and GC-MS/MS

The quantitation of the hop varietal thiols 4-mercapto-4-methyl-2-pentanone (4MMP), 3-mercapto-1-hexanol (3MH), and 3-mercaptohexylacetate (3MHA) from beer is challenging. This primarily relates to their low concentration (ng/L levels) and their reactivity. Published assays for thiol quantitation from beer include complex and/or time-consuming sample preparation procedures involving manual handling and use reagents that are harmful because they contain mercury. To facilitate thiol analysis from beer, the current article is concerned with the implementation of an automated headspace solid-phase microextraction (HS-SPME) on-fiber derivatization (OFD) approach using 2,3,4,5,6-pentafluorobenzyl bromide followed by gas chromatography-tandem mass spectrometry (GC-MS/MS). Optimization of HS-SPME and MRM conditions was based on a central composite design approach. The final OFD-HS-SPME-GC-MS/MS method yielded limits of quantitation below the sensory thresholds of 4MMP, 3MH, and 3MHA. Method validation and application on beers brewed with German, Australian, and US hops, as well as with added fruits displayed excellent method performance.

Coating of Ca(OH)(2)/gamma-Al2O3 pellets with mesoporous Al2O3 and its application in thermochemical heat storage for CSP plants

Thermochemical heat storage using the Ca(OH)(2) dehydration/hydration reaction for CSP plants requires the development of advanced materials that avoids the breakage shown by conventional CaO pellets. In this work, spherical cores made up of pelletized 60 wt % Ca(OH)(2) and 40% gamma-Al2O3 powders were coated with different mesoporous alumina coverings by means of a dip-coating route, in order to enhance its structural integrity for dehydration/hydration cycles. Three strategies of preparation were devised using as coatings alumina gels containing cetyltrimethylammonium bromide as surfactant in neutral (A2 CS) and acid pH (A4 CS) and without surfactant in neutral medium (A3 CS). In both A2 CS and A4 CS materials, denser alumina coatings were attained originating higher crushing strengths, pointing out the positive role played by the surfactant in the gel makeup. SEM images indicated that A2 presented small alumina grains (0.3-3 mm) highly dispersed over their surface, while over A4 CS, long sheets were appreciated. All the materials exhibited high hydration capacities after 10 cycles (>80%, >800 kJ/kg) at 250 Alpha C, but only A2 CS hold its structural integrity that was preserved even after 20 cycles without loss of its hydration capacity. Micro-solvation and counter ion effects on ionic reactions: Activation of potassium fluoride with 18-crown-6 and tert-butanol in aprotic solvents

Ionic reactions have an important solvent effect and the first solvation shell makes the most important contribution for determining the reactivity. In the case of nucleophilic fluorination with potassium fluoride, the solubilization of the salt is another aspect which needs be considered. In the present work, the effect of adding stoichiometric quantity of 18-crown-6 and the bulky alcohol tert-butanol to the reaction of potassium fluoride with alkyl bromide in toluene and acetonitrile solvents was investigated by theoretical methods as molecular dynamics and electronic structure calculations, as well as by experiments. The calculations indicate that the fluoride ion in the IT (18-crown-6) complex interacts with tern-butanol molecules forming aggregates with one (acetonitrite solvent) and three (toluene solvent) tert-butanol molecules. The calculations predict that formation of these aggregates increase the solubilization of KF and produces and important rate acceleration effect in both acetonitrile and toluene solvents, which is experimentally observed. In the concentration around 0.75 mol L-1 of the tert-butanol, the reaction rate is proportional to the alcohol concentration and the rate of alkyl halide decay becomes 10 times higher than the rate without this alcohol. The present results point out that both the interactions of KF with crown ether and tert-butanol molecules have a synergic effect, increasing its reactivity towards S(N)2 reactions. Understanding the Chemistry and Sources of Precipitation Ions in the mid-Brahmaputra Valley of Northeastern India

The chemistry of rainwater over mid-Brahmaputra Valley was studied for three consecutive years (2012-2014; n = 285). The samples were analyzed for major chemical parameters viz. pH, electrical conductivity (EC), and ions (SO42-, NO3-, Cl-, F-, Br-, Ca2+, NH4+, Mg2+, Na+, K+, and Li+), organic acids (HCOO- and CH3COO-) and dissolved organic carbon (DOC). The mean pH for the entire study period was found to be 5.66, which ranged from 4.51 to 7.68, and the volume weighted (VW) mean pH was found to be 5.16. Over 55% of the samples showed pH between 5 and 6, and a few samples had pH 5. Ionic concentration followed the order NH4+ Ca2+ > SO42- > NO3- > Cl- > Na+ > K+ > Mg2+ > H+ > HCO3- > Br- > F- > Li+, indicating dominance of alkaline ions over acidic ions such that 94% of mineral acid was neutralized. The secondary ions, NH4+, SO42-, and NO3-, showed high wet deposition fluxes. The chemistry exhibits explicit seasonality. The airmass clusters of monsoon and non-monsoon seasons, and the associated chemistry varied, which showed influence of long-range transport. The interspecies correlations varied between the monsoon and non-monsoon time samples meaning variation in the source strengths of the contribution sources of the chemical species of the rainwater. Positive Matrix Factorization (PMF) was applied to the data which extracted six factors that explained the sources and chemistry of the rainwater constituents which are of sea, agriculture, coal burning, biomass burning, and secondary origin.

PLCG2 promotes hepatocyte proliferation in vitro via NF-kappa B and ERK pathway by targeting bcl2, myc and ccnd1

Phospholipase C gamma 2 (PLCG2) has been implicated in the regulation of cell proliferation, transformation, and tumor growth. In this study, we investigate the mechanism of PLCG2 action using a short interference RNA (siRNA) method. The effects of PLCG2 on rat liver BRL-3A cells treated siRNA were studied by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT assay), bromodeoxyuridine (BrdU) labelling assay, flow cytometry method (FCM), quantitative real-time polymerase chain reaction (qRT-PCR) and western blot. The results showed when PLCG2 was reduced, cell vitality and proliferation rate were significantly decreased (p < .05 vs. control). FCM analysis showed that the number of cell division phase (G2 + M) was declined (p < .05 vs. control). RT-PCR and western blot revealed that the expression of signalling related genes NF-kappa B, FOS, JUN and ELK, target genes BCL2, CCNB1 and CCND1 were remarkably down-regulated in cells treated with PLCG2 siRNAs. Based on these results, we conclude PLCG2 plays an important role in rat liver cell proliferation via ERK and NF-kappa B pathway by regulating the expression of BCl2, MYC and CCND1.
