Overexpression of miR-129-5p Mitigates Sepsis-Induced Acute Lung Injury by Targeting High Mobility Group Box 1

Background: MicroRNAs are dysregulated in sepsis. Acute lung injury is a progressive syndrome during sepsis. However, the role of miR-129-5p in the development of acute lung injury induced by sepsis remains unclear. Methods: The acute lung injury of sepsis model was established by cecal ligation puncture (CLP)-treated mice and lipopolysaccharide (LPS)-treated murine alveolar epithelial cell line (MLE)-12 cells. The lung injury in vivo was investigated by hematoxylin and eosin staining, terminal dexynucleotidyl transferase (TdT)-mediated dUTP nick end labeling staining, enzyme-linked immunosorbent assay, lung wet-to-dry weight ratio, and myeloperoxidase activity. The lung injury in vitro was evaluated by 3-(4,5-dimethyl-2-thiazolyl)-2, 5-diphenyl-2-H-tetrazolium bromide, flow cytometry, and enzyme-linked immunosorbent assay. The expression levels of miR-129-5p and high mobility group box 1 (HMGB1) were measured by quantitative real-time polymerase chain reaction and Western blot. The association between miR-129-5p and HMGB1 was validated by luciferase assay and RNA immunoprecipitation. Results: The expression of miR-129-5p was decreased in CLP model and LPS-treated MLE-12 cells. Overexpression of miR-129-5p attenuated inflammatory response, apoptosis, lung wet/dry weight ratio, and myeloperoxidase activity induced by CLP surgery in vivo. Moreover, addition of miR-129-5p increased cell viability and suppressed cell apoptosis and inflammatory response in vitro. HMGB1 as a target of miR-129-5p alleviated miR-129-5p emediated injury suppression in LPS-treated MLE-12 cells. Conclusions: miR-129-5p protects against sepsis-induced acute lung injury by decreasing HMGB1 expression, providing new target for sepsis treatment. (C) 2020 Elsevier Inc. All rights reserved.

Accelerating Effect of DMAP on CuI Catalyzed Buchwald-Hartwig C-N Coupling: Mechanistic Insight to the Reaction Pathway

An efficient methodology has been developed for C-N cross-coupling reaction between aryl halide and amine using a catalytic system comprised of CuI and N,N-dimethyl amino pyridine (DMAP). Coupling reactions were carried out in DMSO at 120 degrees C in presence of 5 mol% CuI, 20 mol% DMAP and 2.5 equivalent of KOH. Addition of DMAP was found to be beneficial in accelerating the rate of reaction significantly. Both aryl iodides and aryl bromides could be successfully coupled with different primary and secondary amines to furnish the desired cross-coupling product in high yield. A copper complex [Cu(DMAP)(4)I]I made by combining CuI and DMAP was found to produce superior result for the Buchwlad-Hartwig cross coupling reaction. The use of 2 mol% of the copper complex is sufficient to produce high yield of the product.

Biodegradable CA/CPB electrospun nanofibers for efficient retention of airborne nanoparticles

The increase of the industrialization process brought the growth of pollutant emissions into the atmosphere. At the same time, the demand for advances in aerosol filtration is evolving towards more sustainable technologies. Electrospinning is gaining notoriety, once it enables to produce polymeric nanofibers with different additives and also the obtaining of small pore sizes and fiber diameters; desirable features for air filtration materials. Therefore, this work aims to evaluate the filtration performance of cellulose acetate (CA) nanofibers and cationic surfactant cetylpyridinium bromide (CPB) produced by electrospinning technique for retention of aerosol nanoparticles. The pressure drop and collection efficiency measurements of sodium chloride (NaCl) aerosol particles (diameters from 7 to 300 nm) were performed using Scanning Mobility Particle Sizer (SMPS). The average diameter of the electrospun nanofibers used was 239 nm, ranging from 113 to 398 nm. Experimental results indicated that the nanofibers showed good permeability (10(-11) m(2)) and high-efficiency filtration for aerosol nanoparticles (about 100 %), which can include black carbon (BC) and the new coronavirus. The pressure drop was 1.8 kPa at 1.6 cm s(-1), which is similar to reported for some high-efficiency nanofiber filters. In addition, it also retains BC particles present in air, which was about 90 % for 375 nm and about 60 % for the 880 nm wavelength. Finally, this research provided information for future designs of indoor air filters and filter media for facial masks with renewable, non-toxic biodegradable, and potential antibacterial characteristics. Potential use of 13-mer peptides based on phospholipase and oligoarginine as leishmanicidal agents

Phospholipase A(2) toxins present in snake venoms interact with biological membranes and serve as structural models for the design of small peptides with anticancer, antibacterial and antiparasitic properties. Oligoarginine peptides are capable of increasing cell membrane permeability (cell penetrating peptides), and for this reason are interesting delivery systems for compounds of pharmacological interest. Inspired by these two families of bioactive molecules, we have synthesized two 13-mer peptides as potential antileishmanial leads gaining insights into structural features useful for the future design of more potent peptides. The peptides included p-Acl, reproducing a natural segment of a Lys49 PLA(2) from Agkistrodon contortrix laticinctus snake venom, and its p-AclR7 analogue where all seven lysine residues were replaced by arginines. Both peptides were active against promastigote and amastigote forms of Leishmania (L.) amazonensis and L. (L.) infantum, while displaying low cytotoxicity for primary murine macrophages. Spectrofluorimetric studies suggest that permeabilization of the parasite’s cell membrane is the probable mechanism of action of these biomolecules. Relevantly, the engineered peptide p-AclR7 was more active in both life stages of Leishmania and induced higher rates of ethidium bromide incorporation than its native template p-Acl. Taken together, the results suggest that short peptides based on phospholipase toxins are potential scaffolds for development of antileishmanial candidates. Moreover, specific amino acid substitutions, such those herein employed, may enhance the antiparasitic action of these cationic peptides, encouraging their future biomedical applications.

A dual functional Janus membrane combining superwettability with electrostatic force for controllable anionic/cationic emulsion separation and in situ surfactant removal

In the oil/water emulsion treatment field, the separation of dispersed tiny oil droplets and the removal of surfactants are both significant in order to obtain purified water. Superwetting materials have proved to be an ideal solution for stabilized emulsion separation. However, most of the previous studies only focus on the separation of oil from water while neglecting the removal of surfactants. With the purpose of realizing in situ emulsion separation and surfactant removal, a dual functional Janus membrane is designed inspired by mussel-inspired chemistry and an EDC/NHS coupling reaction. After modification, abundant amino groups and carboxyl groups are decorated on two sides of the microfiltration membrane substrate, respectively (denoted as the -NH2 side and -COOH side). Combining special wettability with electrostatic interaction, the as-prepared membrane is able to separate stabilized oil-in-water emulsions and remove anionic/cationic surfactants simultaneously. The -NH2 side is used for the treatment of anionic emulsions with the surfactant sodium dodecyl sulfate (SDS), while the -COOH side is used for the treatment of cationic emulsions with the surfactant hexadecyl trimethyl ammonium bromide (CTAB). More importantly, the separation efficiencies are all above 99.5% and the surfactant removal efficiencies are up to 95.7%. To our knowledge, this is the first time to treat both dispersed oil droplets and surfactants using superwetting materials. The excellent recyclability and stability of the Janus material make it a potential candidate for application in the industrial field.

Fe3O4@Au/reduced graphene oxide nanostructures: Combinatorial effects of radiotherapy and photothermal therapy on oral squamous carcinoma KB cell line

Combination therapy including radiotherapy (RT) and photothermal therapy (PTT), as an alternative treatment for cancer treatment, recently has generated substantial interest. Graphene-based nanocarbons and gold content metallic nanoparticles, as one of the most attractive materials with their extraordinary physical and chemical properties, are extremely useful in this regard. We herein reported the effects of combination of cancer therapy including RT (doses of 2 and 4 Gy) and PTT (808 nm laser irradiation, NIR irradiation) as radio-photothermal therapy (RPTT) of KB oral squamous carcinoma cell line in the presence of Fe3O4@Au/reduced graphene oxide (rGO) nanostructures (NSs) at different concentrations. Fe3O4@Au/rGO NSs with different rGO contents have been synthesized by hydrothermal reaction method. They characterized by XRD, FE-SEM, TEM, EDS, FTIR and TGA. Cell viability for cytotoxicity, RT, PTT and RPTT were studied by MTT (3[4,5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide) assay. High photothermal conversion efficiency of Fe3O4@Au/rGO NSs reaches to 61%. The resulting Fe3O4@Au NPs with approximate size of about 10-60 nm covered the rGO nanosheets with 40 wt% rGO at concentration of 20 mu g ml(-1), exhibited satisfactory cytotoxicity. They provided significant cell destruction under RT, PIT and specially RPTT in dose of 4 Gy. Furthermore, they have good biocompatibility on the healthy cells. Our results show that Fe3O4@Au/rGO NSs integrated with radiosensitization, photothermal therapy and their combination promising for nanomedicine and clinical applications.

Direct Z-Scheme Heterojunction of Semicoherent FAPbBr(3)/Bi2WO6 Interface for Photoredox Reaction with Large Driving Force

Metal halide perovskites with direct band gap and strong light absorption are promising materials for harvesting solar energy; however, their relatively narrow band gap limits their redox ability when used as a photocatalyst. Adding a second semiconductor component with the appropriate band structure offsets can generate a Z-scheme photocatalytic system, taking full advantage of the perovskite’s intrinsic properties. In this work, we develop a direct Z-scheme photocatalyst based on formamidinium lead bromide and bismuth tungstate (FAPbBr(3)/Bi2WO6) with strong redox ability for artificial solar-to-chemical energy conversion. With desirable band offsets and strong joint redox potential, the dual photocatalyst is shown to form a semicoherent heterointerface. Ultrafast transient infrared absorption studies employing selective excitation reveal synergetic photocarrier dynamics and demonstrate Z-scheme charge transfer mechanisms. Under simulated solar irradiation, a large driving force photoredox reaction (similar to 2.57 eV) of CO2 reduction coupled with benzyl alcohol oxidation to benzaldehyde is achieved on the Z-scheme FAPbBr(3)/Bi2WO6 photocatalyst, harnessing the full synergetic potential of the combined system.
