The aggregation and micellization of ionic surfactants in aqueous solution detected using surface-confined redox and ion-pairing reactions

This article describes the use of a redox-induced ion-pairing reaction confined to an electrode surface to detect the micellar aggregation of ionic surfactants in water. Cyclic voltammetry characterizes the Faradaic response of self-assembled monolayers (SAMs) of ferrocenyldodecanethiolate on gold in aqueous solutions of the sodium salts of the anionic surfactants n-decyl sulfate, n-decyl sulfonate, and taurodeoxycholate, the bromide salt of the cationic surfactant decyltrimethylammonium, and the sodium salt of the azo dye Orange II. These electrolytes present different aggregation behaviors in water. The cyclic voltammograms indicate that the surfactant and dye anions and the bromide counteranion of the cationic decyltrimethylammonium pair with the electrogenerated SAM-bound ferrocenium. The voltammetric signatures are sensitive to the concentration and aggregation state of the ions in solution. The concentration dependence of the apparent redox potential (E degrees'(SAM)) presents the same features observed by potentiometry using surfactant ion-selective or bromide electrodes, indicating that E degrees'(SAM) tracks the activity of the unaggregated or free surfactant, dye or bromide anions in solution. The formation of micellar aggregates above the critical micelle concentration (cmc) causes the free anion activity to deviate from the molar concentration of added surfactant, resulting in a break in the plot of E degrees'(SAM) versus the logarithm of the surfactant concentration. The quantity of surfactant anions associated to the oxidized SAM increases with the solute concentration and plateaus out at concentrations > cmc. MiR-16 inhibits hepatocellular carcinoma progression by targeting FEAT through NF-kappa B signaling pathway

OBJECTIVE: MicroRNA-16 (miR-16) expression has been proved to take part in the initiation and development of several cancers, including hepatocellular carcinoma (HCC). However, its role and its molecular mechanism in HCC cells remain unclear. Our study aimed to elucidate miR-16 probable role and potential mechanism in HCC cells. PATIENTS AND METHODS: MiR-16 expression in HCC was measured by Real Time-Polymerase Chain Reaction (RT-PCR). MiR-16 mimic or inhibitor was applied to increase or decrease miR-16 expression in Huh7 cells separately. The cell viability was measured by MTT (3-(4,5-dimethylthiazol-2-yl)-2.5-diphenyl tetrazolium bromide). The invaded cells and migrated cells were detected by the transwell assay. The epithelial-mesenchymal transition (EMT) and the nuclear factor-kappa B (NF-kappa B) were performed using Western blot. The tumor growth was measured via xenograft tumor formation assay. Moreover, bioinformatical methods and luciferase reporter assay were carried out to confirm the miR-16 target gene. RESULTS: MiR-16 expression was downregulated in HCC tissues and cells. Furthermore, the increasing miR-16 expression was suppressed. whereas the decreasing miR-16 expression promoted cell proliferation, invasion, and migration in Huh7 cells. Moreover, miR-16 targeted FEAT in regulating HCC progression. FEAT was associated with a poor prognosis of HCC patients. Especially. miR-16 suppressed EMT and NF-kappa B pathway in HCC and inhibited the tumor growth in vivo. CONCLUSIONS: We stated that miR-16 suppressed HCC cell progression by targeting FEAT and inhibiting EMT and NF-kappa B pathway. MiR-16 may be clinically utilized as a factor for the clinical diagnosis and prognosis of HCC.

Adenosine 5’-triphosphate (ATP), as a molecular currency for intracellular energy transfer, not only participates in various physiological activities, but also involves in various pathological processes of living cells. Therefore, the establishment of a simple, rapid, sensitive and accurate ATP analysis method is of great significance for the study of its physiological functions. In this work, we rationally design a near-infrared phenylboronic acid group-functionalized dicyanovinyl squaraine (SQ- PBA3), with which a supramolecular assembly was constructed for fluorescent imaging of ATP. In PBS buffer, SQ-PBA3 changes self-assembly when adding cetyltrimethyl ammonium bromide (CTAB) deriving fluorescence response of SQ-PBA3 to ATP with the maximum emission at 700 nm, due to the specificity of phenylboronic acid to diols and the multiple electrostatic interactions between SQ PBA3, ATP and CTAB molecules. Meanwhile, the probe was successfully applied to monitor intracellular ATP level in MCF-7 cells with high sensitivity and selectivity.

Electrospun Nanofibrous Poly (Lactic Acid)/Titanium Dioxide Nanocomposite Membranes for Cutaneous Scar Minimization

Poly (lactic acid) (PLA) has been increasingly used in cutaneous tissue engineering due to its low cost, ease of handling, biodegradability, and biocompatibility, as well as its ability to form composites. However, these polymers possess a structure with nanoporous that mimic the cellular environment. In this study, nanocomposites are prepared using PLA and titanium dioxide (TiO2) (10 and 35%-w/w) nanoparticles that also function as an active anti-scarring agent. The nanocomposites were prepared using an electrospinning technique. Three different solutions were prepared as follows: PLA, 10% PLA/TiO2, and 35% PLA/TiO2 (w/w%). Electrospun PLA and PLA/TiO2 nanocomposites were characterized morphologically, structurally, and chemically using electron scanning microscopy, transmission electron microscopy, goniometry, and X-ray diffraction. L929 fibroblast cells were used for in vitro tests. The cytotoxic effect was evaluated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assays. Versicam (VCAN), biglicam (BIG), interleukin-6 (IL6), interleukin-10 (IL-10), and type-1 collagen (COL1A1) genes were evaluated by RT-qPCR. In vivo tests using Wistar rats were conducted for up to 15 days. Nanofibrous fibers were obtained for all groups that did not contain residual solvents. No cytotoxic effects were observed for up to 168 h. The genes expressed showed the highest values of versican and collagen-1 (p < 0.05) for PLA/TiO2 nanocomposite scaffolds when compared to the control group (cells). Histological images showed that PLA at 10 and 35% w/w led to a discrete inflammatory infiltration and expression of many newly formed vessels, indicating increased metabolic activity of this tissue. To summarize, this study supported the potential of PLA/TiO2 nanocomposites ability to reduce cutaneous scarring in scaffolds. The Use of Activated Micronized Zeolite Clinoptilolite as a Possible Alternative to Antibiotics and Chestnut Extract for the Control of Undifferentiated Calf Diarrhea: An In Vitro and In Vivo Study

Simple Summary Today, zeolite has appeared as an interesting alternative for the symptomatic treatment of acute diarrhea. Therefore, this study aimed to investigate the properties of activated micronized (five microns) zeolite clinoptilolite (MZC) from Transylvania, Romania, first by testing it in vitro and then in vivo on calves with diarrhea. To assess the toxic potential of MZC, we performed a cell cytotoxicity assay on cells of bovine origin, while its antimicrobial activity was investigated on Escherichia coli. The uncontrolled in vivo study was carried out over 8 days on a fattening farm, with some 650 calves. Selected calves were randomly assigned to 4 groups of 20 individuals. Several combinations between activated MZC, chestnut extract, and oxytetracycline were tested. At the beginning of the study, all calves had diarrhea, while at the end of the study, the prevalence of diarrhea was significantly lower in all four groups (p < 0.001), including the ones treated with MZC. Due to its low cytotoxicity on the intestinal cells and with regards to the results we obtained in vivo, MZC could represent an alternative method to reducing the amount of antimicrobials needed for the symptomatic treatment of diarrhea in calves, therefore contributing to the reduction of the antimicrobial resistance phenomenon. Today, zeolite appears as an interesting alternative for the symptomatic treatment of acute diarrhea. Synthesis and anti-methicillin-resistant Staphylococcus aureus activity of 5,7-dibromo-2-benzoylbenzofurans alone and in combination with antibiotics

A series of 5,7-dibromo-2-benzoylbenzofurans were synthesized by the Rap-Stoermer condensation of 5,7-dibromosalicylaldehyde with diverse phenacyl bromides and evaluated for in-vitro antibacterial activities against methicillin-sensitive Staphylococcus aureus (MSSA) ATCC 29213, methicillin-resistant Staphylococcus aureus (MRSA) ATCC 43300, and MRSA ATCC 33591 by agar dilution method. The synergistic effects were determined by using the agar dilution checkerboard assay. The derivatives bearing carboxylic acid functional groups exhibited reasonable activity against MRSA strains with the best MIC = 32 mu g/mL (9b, 9d). Moreover, the additive or synergistic interactions against MRSA strains was observed in six combinations (1b + cefuroxime/gentamicin, 1c + ciprofloxacin/gentamicin, 9b + gentamicin, and 9c + ciprofloxacin) with the fractional inhibitory concentration index (FICI) values in the range of 0.375-1.0. Significantly, the MICs of these antibiotics were reduced 2-4-fold. The results of the MTT assay illustrated the low mammalian cell cytotoxicity of these potent compounds.

Seed-Adsorbate Interactions as the Key of Heterogeneous Butanol and Diethylene Glycol Nucleation on Ammonium Bisulfate and Tetramethylammonium Bromide

Condensation particle counter (CPC) instruments are commonly used to detect atmospheric nanoparticles. They operate on the basis of condensing an organic working fluid on the nanoparticle seeds to grow the particles to a detectable size, and at the size of few nanometers, their efficiency depends on how well the working fluid interacts with the seeds under the measurement conditions. This study models the first steps of heterogeneous nucleation of two working fluids commonly used in CPCs (diethylene glycol (DEG) and n-butanol) onto two positively charged seeds, ammonium bisulfate and tetramethylammonium bromide. The nucleation process is modeled on a molecular level using a combination of systematic configurational sampling and density functional theory (DFT). We take into account the conformational flexibility of DEG and n-butanol and determine the key factors that can improve the efficiency of nanoparticle measurements by CPCs. The results show that hydrogen bonding between the seed and the working fluid molecules is central to the adsorption of the first DEG/n-butanol molecules onto the seeds. However, intermolecular hydrogen bonding between the adsorbed molecules can also enhance the nucleation process for the weakly adsorbing vapor molecules. Accordingly, the heterogeneous nucleation probability is higher for working fluid-nanoparticle combinations with a higher potential for hydrogen bonding; in this case, DEG and ammonium bisulfate. Moreover, conformational analysis and methodology evaluations indicate that the consideration of adsorbate conformers and step-wise addition of the vapor molecules to the seeds is not essential for qualitative modeling of heterogeneous nucleation systems, at least for systems where the adsorbate and seed chemical properties are clearly different. This is the first molecular-level modeling study reporting detailed chemical reasons for experimentally observed seed and working fluid preferences in CPCs and reproducing the experimental observations. Our presented approach can be likely used for predicting preferences in similar nucleating systems.
