A protocol for the generation and precise control of the sulfonyl radical from dimethylsulfamoyl chloride was reported. Different kinds of alkyl sulfonates and sulfonamides were efficiently and selectively obtained under redox-neutral conditions by controlling the reactivity of photocatalysts. A mechanism for the generation of sulfamoyl and sulfonate radicals from dimethylsulfamoyl chloride and the followed sulfonation and sulfonamidation were proposed. This reaction system offers a distinct advantage over traditional oxidation or reduction sulfonation methods in not requiring the use of sulfur dioxide, sulfonic acid, a stoichiometric reductant, or a stoichiometric oxidant. Thus, it should be of practical value for the late-stage functionalization of natural products or medicines.

Zhang, Mingjun;Ding, Xin;Kang, Jin;Gao, Yongyue;Wang, Ziwen;Wang, Qingmin research published 《 Marine Natural Product for Pesticide Candidate: Pulmonarin Alkaloids as Novel Antiviral and Anti-Phytopathogenic-Fungus Agents》, the research content is summarized as follows. Plant diseases are seriously endangering agricultural production The emergence of drug resistance has brought great challenges to the prevention and control of plant diseases. There is an urgent need for the emergence of new drug candidates. In this work, we achieved the efficient synthesis of pulmonarins A and B in 64% and 59% overall yield, resp. Pulmonarins A and B were found to have good antiviral activities against tobacco mosaic virus (TMV) for the first time. A series of pulmonarin derivatives were designed, synthesized, and evaluated for their antiviral and fungicidal activities systematically. Most compounds displayed higher anti-TMV activities than com. ribavirin. Compounds Pulmonarin B, -Benzyl-2-((3,5-dibromo-4-methoxybenzoyl)oxy)-N,N-dimethylethan-1-aminium Bromide (I), and 2-((4-(Allyloxy)-3,5-dibromobenzoyl)oxy)-N,N,N-trimethylethan-1-aminium Iodide with better inactivation effects than ningnanmycin emerged as new antiviral candidates. We selected I for further antiviral mechanism research, which revealed that it could inhibit virus assembly by interacting with TMV coat protein (CP). The mol. docking results further confirmed that these compounds could interact with CP through hydrogen bonding. These compounds also displayed broad spectrum fungicidal activities. Especially compound 2-(2-(3,5-Dibromo-4-methoxyphenyl)acetoxy)-N,N,N-trimethylethan-1-aminium Iodide with prominent antifungal activity emerged as a new fungicidal candidate for further research. The current work provides a reference for understanding the application of pulmonarin alkaloids in plant protection.

Reductive arylation of secondary diarylphosphine oxides Ar2P(O)H was achieved by one-pot reaction with DIBAL-H and arylammonium salts [Ar1NMe3][OTf] in the presence of KOtBu base and transition metal-free conditions affording triarylphosphines Ar2PAr1 with high yields. A nucleophilic aromatic substitution (SNAr) reaction that allowed transition-metal-free C-P bond construction via C-N bond cleavage was developed. The coupling between aryltrimethylammonium salts and secondary phosphines from the in situ reduction of diarylphosphine oxides led to the formation of diverse triarylphosphines with various functional groups. This one-pot process was not only a pertinent SNAr precedent but also a favorable transition-metal-free alternative for C-P coupling.

Zhang, Jun;Zheng, Mengmeng;Yan, Jiayan;Deng, Zixin;Zhu, Dongqing;Qu, Xudong research published 《 A Permissive Medium Chain Acyl-CoA Carboxylase Enables the Efficient Biosynthesis of Extender Units for Engineering Polyketide Carbon Scaffolds》, the research content is summarized as follows. The selective incorporation of new-to-nature extender units into polyketide synthesis is a highly effective way to engineer their carbon scaffolds. Currently, most atypical extender units are biosynthesized via reductive carboxylation of α,β-unsaturated thioesters catalyzed by crotonyl-CoA reductase/carboxylases or thioesterification of malonates by malonyl-CoA ligases followed by epimerase catalyzed enantiomerization. In this study, we identified an unusual β-subunit (Arm13) of the acyl-CoA carboxylase (ACCase) from the biosynthesis of armeniaspirols. This β-subunit is permissive to the α- and ε-subunits of propionyl-CoA carboxylase to form a fully active ACCase. Distinct from the other regular ACCases in substrate specificity, this ACCase can directly carboxylate medium chain acyl-CoAs ranging from C6 to C9 either with or without terminal substituents, e.g., alkyne or Ph groups, to produce corresponding alkylmalonyl-CoAs with high catalytic efficiency. By harnessing the power of this ACCase in extender unit biosynthesis, we introduced structural variation into the carbon scaffold of armeniaspirols by feeding corresponding carboxylate precursors. These findings not only enrich the knowledge of the medium chain-specific ACCases but also provide an important biocatalyst for diversifying building blocks, which will greatly facilitate polyketide engineering.

Zhang, Jitan;Xu, Qiaoqiao;Fan, Jian;Zhou, Lan;Liu, Nannan;Zhu, Li;Wu, Jiaping;Xie, Meihua research published 《 Pd(II)-Catalyzed enantioconvergent twofold C-H annulation to access atropisomeric aldehydes: a platform for diversity-oriented-synthesis》, the research content is summarized as follows. Herein, the first Pd(II)-catalyzed atroposelective dual C-H annulative strategy for the assembly of axially chiral aldehyde frameworks such as I [R = Me, Et, i-Pr] using com. available amino acid as the transient auxiliary and chiral pool was presented. This reaction accommodated a broad substrate scope to give atropisomeric aldehydes bearing both C-C and N-C chiral axes in good yields (up to 95%) with excellent enantioinduction (up to 99%). The utility of this synthetic methodol. was testified by various practical late-stage transformations, thereby accomplishing diversity-oriented synthesis of structurally diverse biaryl atropisomers and several functionalized axially chiral species such as bifunctional organocatalysts. Moreover, a series of mechanistic studies had provided more details for this catalytic transformation.
