An efficient, mild, and green method was developed for the synthesis of indeno[1,2-b]quinoxaline derivatives via o-phenylenediamine (OPD) and 2-indanone derivatives utilizing β-cyclodextrin (β-CD) as the supramol. catalyst. The reaction can be carried out in water and in a solid state at room temperature β-CD can also catalyze the reaction of indan-1,2-dione with OPD with a high degree of efficiency. Compared to the reported methods, this procedure is milder, simpler, and less toxic, making it an eco-friendly alternative. In addition, the β-CD can be recovered and reused without the loss of activity.

An efficient one-pot reaction for the synthesis of oxoaporphine alkaloids has been developed. Twenty-three compounds of oxoaporphine alkaloids were prepared and assessed for their antitumor activities. Most compounds inhibited the growth of T-24 tumor cells in vitro. Particularly, I displayed the most potent activity with an IC50 value of 0.5μM, which was 19-fold more potent than the parent compound 4. The substitution at C3-position of oxoaporphine core by -NO2 significantly enhanced the anticancer activity. Mechanism studies indicated that II and I induced cell cycle arrest at G2/M phase; in contrast, III induced cell cycle arrest at the S phase. Increase of mitochondrial ROS/Ca2+ and decrease of MMP, accompanied by activation of caspase-3/9, were observed in T-24 cells after exposure to compounds I, II and III, suggesting that the mitochondrial pathway was involved in the induced apoptosis. Moreover, compound I effectively inhibited tumor growth in a mouse xenograft model bearing T-24.

Aryl sulfides are in great demands in drugs and materials sciences. To avoid using nucleophilic and noxious thiols, many efforts have been focused on exploring novel sulfide resources. Herein, a reductive Pd-catalyzed, Ni-mediated method to synthesize aryl sulfides via a sulfide transfer reaction is developed. The utility and scope of this reaction is exemplified by various aryl electrophiles and aryl sulfides. Mechanistic studies reveal two competing catalytic cycles of sulfide transfer and aryl transfer in this reaction, where the former one is favored over the later one because of the large energy barrier difference during the transmetalation. Moreover, two important chems. are late-stage functionalized by this method, exhibiting the potential applications in drugs and materials science.

Liang, Tao;Zhou, Yi;Elhassan, Reham M.;Hou, Xuben;Yang, Xinying;Fang, Hao research published 《 HDAC-Bax Multiple Ligands Enhance Bax-Dependent Apoptosis in HeLa Cells》, the research content is summarized as follows. Inspired by the synergistic effect of BTSA1 (a Bax activator) and SAHA (a histone deacetylase (HDAC) inhibitor) in HeLa cell growth suppression, a series of novel HDAC-Bax multiple ligands were designed rationally. Compound 23, which possesses similar HDAC inhibitory activity relative to SAHA and Bax affinity comparable to BTSA1, exhibits a superior growth suppression against HeLa cells, and its antiproliferative activities are 15-fold and 3-fold higher than BTSA1 and SAHA, resp. The better antiproliferative activity and lower cytotoxicity of compound 23(I) indicated that our HDAC-Bax multiple ligand design strategy achieved success. Further studies suggested that compound 23 could enhance Bax-dependent apoptosis by upregulating Bax, followed by inducing the conformational activation of Bax. To our knowledge, we first report HDAC-Bax multiple ligands and demonstrate a new paradigm for the treatment of solid tumors by enhancing Bax-dependent apoptosis.

Li, YiXiang;Wu, Yong;Qin, ZhengSheng;Wang, Gang;Wu, Yuan;Wang, DongDong;Zhang, YiFan;Wang, XinYe;Wu, ChuanMing;Dong, HuanLi research published 《 Substitution effect on solid parking motif and luminescence of diphenylfuro[2,3-b]quinoxaline isomers》, the research content is summarized as follows. The organic mols. combining light-emitting and charge-transporting property are highly expected. We designed and synthesized a pair of isomers, 2,6-diphenylfuro[2,3-b]quinoxaline (26dPFQ) and 2,7-diphenylfuro[2,3-b]quinoxaline (27dPFQ), and investigated effect of changing substituent position on mol. packing motif, conducting and luminescence feature of resulting materials. In CH2Cl2 solution, both isomers, 26dPFQ and 27dPFQ, emit deep blue light with a wavelength of 422 nm and shows photoluminance quantum yield (PLQY) of 58.9% and 48.7%, resp. In crystal, the 26dPFQ employs H-type packing motif and retains blue florescent emission with low PLQY (10.0%) while 27dPFQ adopts J-aggregated packing motif and shows enhanced green fluorescent emission with a PLQY of 55.0%. The organic light-emitting devices for both emitters show deep blue emission, the EL peak at 410 nm for 26dPFQ and 424 nm for 27dPFQ, and resp. presents the maximum EQE of 1.2% and 1.0%. The 26dPFQ shows better charge transport capability over 27dPFQ. Our results suggested 2,6-substitution on furo[2,3-b]quinoxaline core are expected to afford such dual functional materials.

Li, Yigao;Ren, Xiaohuang;Chen, Yi;Zhu, Xinju;Hao, Xin-Qi;Song, Mao-Ping research published 《 Fe(III)-Catalyzed N-Amidomethylation of Secondary and Primary Anilines with TosMIC》, the research content is summarized as follows. A Fe(III)-catalyzed N-amidomethylation of secondary and primary anilines with p-toluenesulfonylmethyl isocyanide (TosMIC) in water was described. TosMIC played dual roles as the source of methylene as well as an amidating reagent to form α-amino amides R1N(R2)CH2C(O)NHR3 [R1 = Ph, 2-MeC6H4, 4-FC6H4, etc.; R2 = H, Me, Bn, etc.; R3 = t-Bu, 4-MeC6H4SO2CH2, 4-ClC6H4SO2CH2] in this multicomponent reaction. The combination of TosMIC and other isocyanides was also investigated to give the desired products in acceptable yields. The current protocol features used of iron catalyst and nontoxic media, broad substrate scope, mild conditions and operational simplicity.
