A new series of acridine-9-carboxamide-1,2,3-triazole derivatives I [R = H, 2-Me, 4-O2N, etc.] were designed, synthesized and evaluated as novel α-glucosidase inhibitors. Compounds I were designed by combination of effective moieties from potent α-glucosidase inhibitors. Most of the synthesized compounds I were more potent than standard inhibitor acarbose. Among the synthesized compounds, the most potent compounds were I [R = 3-Br, 4-Br, H] with IC50 values of 120.2 ± 1.0, 151.1 ± 1.4, and 157.6 ± 1.6μM, resp. (IC50 value of acarbose = 750.0 ± 10.0μM). Docking study of the most potent compounds I [R = H, 3-Br, 4-Br] reported that these compounds formed stable complexes with α-glucosidase active site. Anti-α-amylase assay of compounds I [R = H, 3-Br, 4-Br] was performed and no activity was observed, meanwhile in-vitro cytotoxicity assay of the latter compounds revealed that these compounds were not cytotoxic toward human normal (HDF) and cancer (MCF-7) cell lines. ADME and toxicity prediction of compounds I [R = H, 3-Br, 4-Br] were also performed.

A new series of 1,2,3-triazole-(thio)barbituric acid hybrids 8a-n was designed and synthesized on the basis of potent pharmacophores with urease inhibitory activity. Therefore, these compounds were evaluated against Helicobacter pylori urease. The obtained result demonstrated that all the synthesized compounds, 8a-n, were more potent than the standard urease inhibitor, hydroxyurea. Moreover, among them, compounds 8a, 8c-e, 8g,h, and 8k,l exhibited higher urease inhibitory activities than the other standard inhibitor used: thiourea. Docking studies were performed with the synthesized compounds Furthermore, mol. dynamic simulation of the most potent compounds, 8e and 8l, showed that these compounds interacted with the conserved residues Cys592 and His593, which belong to the active site flap and are essential for enzymic activity. These interactions have two consequences: (a) blocking the movement of a flap at the entrance of the active site channel and (b) stabilizing the closed active site flap conformation, which significantly reduces the catalytic activity of urease. Calculation of the physicochem. and topol. properties of the synthesized compounds 8a-n predicted that all these compounds can be orally active. The ADME prediction of compounds 8a-n was also performed.

Asano, Shota;Itto-Nakama, Kaori;Arimoto, Hirokazu research published 《 Carbamate as an accelerating group in intermolecular Pauson-Khand reaction》, the research content is summarized as follows. The Pauson-Khand reaction (PKR) is a powerful means for the construction of cyclopentenones. However, its applications are limited to the intramol. version of this reaction because poor yield and regioselectivity are often the major problems in intermol. PKR. Herein, it was shown that a carbamate moiety in alkene substrate accelerates this intermol. PKR. The reaction of N-4-dimethylaminophenyl O-allyl carbamate with alkyne-cobalt complex gave cyclopentenones in high yields (up to 90%) and regioselectivity (>9:1).

1-Bromo-3,5-dimethoxybenzene (1BDMB) is a synthetic molecule that can be used as an electron acceptor in organic photovoltaic cells. 1BDMB is a salt of the sodium salt of resorcylic acid and 1,3-dibromo-5,5-dimethoxybenzene. It has been shown to have a radical mechanism for the generation of free radicals. The radical mechanism is initiated by light absorption by the ruthenium complex at the center of the molecule which induces photoinduced electron transfer from the ruthenium to 1BDMB. Andrews, Mary Katherine;Liu, Xinyu;Gellman, Samuel H. research published 《 Tailoring Reaction Selectivity by Modulating a Catalytic Diad on a Foldamer Scaffold》, the research content is summarized as follows. Use of a tunable mol. scaffold to align a reactive diad for bifunctional catalysis can reveal relationships between functional group identity and reactivity that might otherwise be impossible to identify. Here we use an α/β-peptide helix to show that an aligned pair of primary amine groups is uniquely competent to catalyze crossed aldol condensations with an aryl aldehyde as the electrophile. Geometrically similar diads in which one amine group is secondary, or both are secondary, are good catalysts for other types of aldol condensations but not those involving an aryl aldehyde. Catalytic efficacy requires β-amino acid residues that are preorganized for helix formation via cyclic constraint. Conventional peptides (exclusively α-amino acid residues) that display the primary amine diad are poor catalysts, which highlights the critical role of the foldamer scaffold.

Andleeb, Hina;Danish, Lubna;Munawar, Shiza;Ahmed, Muhammad Naeem;Khan, Imtiaz;Ali, Hafiz Saqib;Tahir, Muhammad Nawaz;Simpson, Jim;Hameed, Shahid research published 《 Theoretical and computational insight into the supramolecular assemblies of Schiff bases involving hydrogen bonding and C-H…π interactions: Synthesis, X-ray characterization, Hirshfeld surface analysis, anticancer activity and molecular docking analysis》, the research content is summarized as follows. The present study examines the significance of various non-covalent interactions in the supramol. assembly of (E)-1-(1-(4-nitrophenyl)ethylidene)-2-phenylhydrazine 1c and (E)-3-bromo-N’-(1-phenylethylidene)benzohydrazide 2d. The synthesized compounds were fully characterized by spectroscopic methods and single crystal X-ray diffraction anal. The topol. of the supramol. assemblies was controlled by various non-covalent interactions including classical hydrogen bonding, C-H…π and Br… Br interactions which were examined in detail using several theor. methods and DFT calculations The optimized geometric parameters of compounds 1c and 2d were calculated using d. functional theory (DFT/B3LYP) quantum chem. method with the 6-311++G(d,p) basis set using the crystallog. coordinates. Addnl., fragments contributing to the HOMO and LUMO MOs were investigated at the same level of theory. The nature and various types of intermol. interactions in the crystal structures was also investigated by Hirshfeld surface anal. The synthesized Schiff bases were also studied for their potential as drugs and physicochem. properties. Bioevaluation against four cancer cell lines (NCI-H460, NCI-H460/Bcl-2, MDA-MB-231 and MCF-7) showed that compound 1c was a more potent inducer of toxicity compared to 2d. The putative binding modes of the bioactive Schiff bases were investigated using mol. docking tools and the results revealed that both the inhibitors were stabilized in the active pocket of the enzyme via the formation of various interactions with the key amino acid residues.

An, Xiaoying;Bai, Fan;Li, Wenhong;Wang, Lanzhi research published 《 Synthesis of 2,3-diester-2,4-diphenyl-1,5-benzodiazepine compounds via one-pot three-component tandem reactions》, the research content is summarized as follows. In this paper, seven novel 2,3-diester-2,4-diphenyl-1,5-benzodiazepines with imine structure were obtained via one-pot three-component tandem reactions catalyzed by TsOH in ethanol. With substituted 1,2-phenylenediamines, Et benzoylacetate and Et benzoylformate as the raw materials, the tandem reactions undergo two nucleophilic addition-dehydration processes to form the active intermediate mols. containing imine and enamine structures, further undergo carbon-carbon coupling cyclization and hydrogen transfer to yield the target products. A reasonable reaction mechanism is proposed for this three-component tandem reaction. The synthesis has the advantages of environmental friendliness, mild reaction conditions, wide substrate range, etc., which provides a green, efficient and convenient synthetic route for nitrogen-containing heterocyclic compounds
