Messmer, Daniel;Bertran, Oscar;Kissner, Reinhard;Aleman, Carlos;Schluter, A. Dieter research published 《 Main-chain scission of individual macromolecules induced by solvent swelling》, the research content is summarized as follows. We present a comprehensive investigation of main-chain scission processes affecting peripherally charged and neutral members of a class of dendronized polymers (DPs) studied in our laboratory In these thick, sterically highly congested macromols., scission occurs by exposure to solvents, in some cases at room temperature, in others requiring modest heating. Our investigations rely on gel permeation chromatog. and at. force microscopy and are supported by mol. dynamics simulations as well as by ESR spectroscopy. Strikingly, DP main-chain scission depends strongly on two factors: first the solvent, which must be highly polar to induce scission of the DPs, and second the dendritic generation g. In DPs of generations 1 ≤ g ≤ 8, scission occurs readily only for g = 5, no matter whether the polymer is charged or neutral. Much more forcing conditions are required to induce degradation in DPs of g ≠ 5. We propose solvent swelling as the cause for the main-chain scission in these individual polymer mols., explaining in particular the strong dependence on g: g < 5 DPs resemble classical polymers and are accessible to the strongly interacting, polar solvents, whereas g > 5 DPs are essentially closed off to solvent due to their more closely colloidal character. g = 5 DPs mark the transition between these two regimes, bearing strongly sterically congested side chains which are still solvent accessible to some degree. Our results suggest that, even in the absence of structural elements which favor scission such as cross-links, solvent swelling may be a generally applicable mechanochem. trigger. This may be relevant not only for DPs, but also for other types of sterically strongly congested macromols.

Miao, Yupeng;Qian, Naixin;Shi, Lixue;Hu, Fanghao;Min, Wei research published 《 9-Cyanopyronin probe palette for super-multiplexed vibrational imaging》, the research content is summarized as follows. Multiplexed optical imaging provides holistic visualization on a vast number of mol. targets, which has become increasingly essential for understanding complex biol. processes and interactions. Vibrational microscopy has great potential owing to the sharp linewidth of vibrational spectra. In 2017, we demonstrated the coupling between electronic pre-resonant stimulated Raman scattering (epr-SRS) microscopy with a proposed library of 9-cyanopyronin-based dyes, named Manhattan Raman Scattering (MARS). Herein, we develop robust synthetic methodol. to build MARS probes with different core atoms, expansion ring numbers, and stable isotope substitutions. We discover a predictive model to correlate their vibrational frequencies with structures, which guides rational design of MARS dyes with desirable Raman shifts. An expanded library of MARS probes with diverse functionalities is constructed. When coupled with epr-SRS microscopy, these MARS probes allow us to demonstrate not only many versatile labeling modalities but also increased multiplexing capacity. Hence, this work opens up next-generation vibrational imaging with greater utilities.

McDonagh, Anthony W.;McNeil, Brooke L.;Patrick, Brian O.;Ramogida, Caterina F. research published 《 Synthesis and Evaluation of Bifunctional [2.2.2]-Cryptands for Nuclear Medicine Applications》, the research content is summarized as follows. For the first time, synthesis of bifunctional [2.2.2]-cryptands (CRYPT) and demonstration of radiolabeling with lead(II) (Pb2+) isotopes are disclosed herein. The synthesis is convenient and high-yielding and gives access to three distinct bifunctional handles (azide (-N3), isothiocyanate (-NCS), and tetrazine (-Tz)) that can enable the construction of radioimmunoconjugates for targeted and pretargeted therapy. Proof-of-principle CRYPT radiolabeling was successful with lead-203 ([203Pb]Pb2+) and demonstrated complexation efficiency superior to that of DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) and efficiency comparable to that of the current industry standard TCMC (1,4,7,10-tetraaza-1,4,7,10-tetra-(2-carbamoylmethyl)-cyclododecane). In vitro human serum stability assays demonstrated excellent [203Pb]Pb-CRYPT stability over 72 h (91.7 ± 0.56%; n = 3). [203Pb]Pb-CRYPT-radioimmunoconjugates were synthesized from the corresponding CRYPT-immunoconjugate or by conjugating [203Pb]Pb-Tz-CRYPT to transcyclooctene modified trastuzumab (TCO-trastuzumab) via the inverse electron-demand Diels-Alder (IEEDA) reaction. This investigation reveals the potential for CRYPT ligands to become new industry standards for therapeutic and diagnostic radiometals in radiopharmaceutical elaboration.

Man, Yan;Ren, Junan;Li, Bingru;Jin, Xinxin;Pan, Ligang research published 《 A simple, highly sensitive colorimetric immunosensor for the detection of alternariol monomethyl ether in fruit by non-aggregated gold nanoparticles》, the research content is summarized as follows. Alternariol monomethyl ether (AME) is one of the major Alternaria mycotoxins present in a wide range of fruits, vegetables, grains, and their products, and possesses the properties of mutagenicity and carcinogenicity. In this study, a simple, rapid, and highly sensitive colorimetric immunosensor based on magnetic nanoparticles (MNPs) was firstly developed for the detection of AME in fruit by nonaggregated gold nanoparticles (GNPs). AME-BSA-Fe3O4 MNP conjugates and free AME mols. in samples competitively bind with monoclonal antibody (mAb)-GNP conjugates. After magnetic separation, the UV absorbance of the nonaggregated GNP supernatant was measured directly. The absorption intensity was proportional to the concentration of AME in the sample. Carboxyl-group-modified AME, AME-bovine serum albumin (BSA) conjugates, anti-AME mAbs, AME-BSA-Fe3O4 MNP conjugates, and mAb-GNP conjugates were prepared and characterized. The effect of GNP sizes (16, 24, and 40 nm) on the colorimetric determination of AME was studied. Under optimized conditions, the limit of detection and the linear range for AME were 0.16 ng/mL and 0.08-0.48 ng/mL, resp. Moreover, the colorimetric immunosensor developed has lower cross-reactivity with AME analogs. The recoveries of spiked fruits ranged from 80.6% to 90.7%. The colorimetric immunosensor developed provides a promising method for simple, rapid, highly sensitive, and highly specific detection of other mycotoxins in the field of food safety. Overexpression of histone deacetylases (HDACs) are observed in different types of cancers, but histone deacetylase inhibitors (HDACIs) have not shown significant efficacy as monotherapy against solid tumors. Recently, studies demonstrated that it is promising to combine HDACIs with DNA damage agents to improve DNA damage level to gain better effect on treating solid tumor. Harmine has been demonstrated to cause DNA damage by intercalating DNA. Therefore, we designed a series of harmine-based inhibitors targeting HDAC and DNA with multi-target strategy, the most potential compound 27 could bind to DNA and cause DNA damage. Furthermore 27 caused cells apoptosis through p53 signaling pathway, and exhibited significant anti-proliferation effects against HCT-116 cells (IC50 = 1.41 μM). As a DNA damage agent, 27 displayed low toxicity in normal cells. Compound 27 was demonstrated as a dual inhibitor targeting HDAC (HDAC1 IC50 = 0.022 μM and HDAC6 IC50 = 0.45 μM) and DNA, and had the potential in the treatment of solid tumor.

A catalytic method to prepare highly substituted 1,3-dienes from two different alkenes is described using a directed, palladium(II)-mediated C(alkenyl)-H activation strategy. The transformation exhibits broad scope across three synthetically useful substrate classes masked with suitable bidentate auxiliaries (4-pentenoic acids, allylic alcs., and bishomoallylic amines) and tolerates internal nonconjugated alkenes, which have traditionally been a challenging class of substrates in this type of chem. Catalytic turnover is enabled by either MnO2 as the stoichiometric oxidant or co-catalytic Co(OAc)2 and O2 (1 atm). Exptl. and computational studies were performed to elucidate the preference for C(alkenyl)-H activation over other potential pathways. As part of this effort, a structurally unique alkenylpalladium(II) dimer was isolated and characterized.

Liu, Wei;Lavagnino, Marissa N.;Gould, Colin A.;Alcazar, Jesus;MacMillan, David W. C. research published 《 A biomimetic SH2 cross-coupling mechanism for quaternary sp3-carbon formation》, the research content is summarized as follows. Bimol. homolytic substitution (SH2) is an open-shell mechanism that is implicated across a host of biochem. alkylation pathways. Surprisingly, however, this radical substitution manifold has not been generally deployed as a design element in synthetic C-C bond formation. Authors found that the SH2 mechanism can be leveraged to enable a biomimetic sp3-sp3 cross-coupling platform that furnishes quaternary sp3-carbon centers, a long-standing challenge in organic mol. construction. This heteroselective radical-radical coupling uses the capacity of iron porphyrin to readily distinguish between the SH2 bond-forming roles of open-shell primary and tertiary carbons, combined with photocatalysis to generate both radical classes simultaneously from widely abundant functional groups. Mechanistic studies confirm the intermediacy of a primary alkyl-Fe(III) species prior to coupling and provide evidence for the SH2 displacement pathway in the critical quaternary sp3-carbon bond formation step.
