Lewis-Borrell, Luke;Sneha, Mahima;Bhattacherjee, Aditi;Clark, Ian P.;Orr-Ewing, Andrew J. research published 《 Mapping the multi-step mechanism of a photoredox catalyzed atom-transfer radical polymerization reaction by direct observation of the reactive intermediates》, the research content is summarized as follows. The rapid development of new applications of photoredox catalysis has so far outpaced the mechanistic studies important for rational design of new classes of catalysts. Here, we report the use of ultrafast transient absorption spectroscopic methods to reveal both mechanistic and kinetic details of multiple sequential steps involved in an organocatalyzed atom transfer radical polymerization reaction. The polymerization system studied involves a N,N-diaryl dihydrophenazine photocatalyst, a radical initiator (Me 2-bromopropionate) and a monomer (isoprene). Time-resolved spectroscopic measurements spanning sub-picosecond to microseconds (i.e., almost 8 orders of magnitude of time) track the formation and loss of key reactive intermediates. These measurements identify both the excited state of the photocatalyst responsible for electron transfer and the radical intermediates participating in propagation reactions, as well as quantifying their lifetimes. The outcomes connect the properties of N,N-diaryl dihydrophenazine organic photocatalysts with the rates of sequential steps in the catalytic cycle.

Lacivita, Enza;Niso, Mauro;Mastromarino, Margherita;Garcia Silva, Andrea;Resch, Cibell;Zeug, Andre;Loza, Maria I.;Castro, Marian;Ponimaskin, Evgeni;Leopoldo, Marcello research published 《 Knowledge-Based Design of Long-Chain Arylpiperazine Derivatives Targeting Multiple Serotonin Receptors as Potential Candidates for Treatment of Autism Spectrum Disorder》, the research content is summarized as follows. Autism spectrum disorder (ASD) includes a group of neurodevelopmental disorders characterized by core symptoms such as impaired social interaction and communication, repetitive and stereotyped behaviors, and restricted interests. To date, there are no effective treatments for these core symptoms. Several studies have shown that the brain serotonin (5-HT) neurotransmission system is altered in both ASD patients and animal models of the disease. Multiple pieces of evidence suggest that targeting 5-HT receptors may treat the core symptoms of ASD and associated intellectual disabilities. In fact, stimulation of the 5-HT1A receptor reduces repetitive and restricted behaviors; blockade of the 5-HT2A receptor reduces both learning deficits and repetitive behavior, and activation of the 5-HT7 receptor improves cognitive performances and reduces repetitive behavior. On such a basis, we have designed novel arylpiperazine derivatives pursuing unprecedently reported activity profiles: dual 5-HT7/5-HT1A receptor agonist properties and mixed 5-HT7 agonist/5-HT1A agonist/5-HT2A antagonist properties. Seventeen new compounds were synthesized and tested in radioligand binding assay at the target receptors. We have identified the dual 5-HT1AR/5-HT7R agonists 8c and 29 and the mixed 5-HT1AR agonist/5-HT7R agonist/5-HT2AR antagonist 20b. These compounds are metabolically stable in vitro and have suitable central nervous system druglike properties.

Herein, the first highly potent 5-HT6/5-HT2A receptor (5-HT6/5-HT2AR) dual antagonists in a group of 1,3,5-triazine compounds have been discovered as a result of an exit beyond the hydrophobic feature of the pharmacophore model for 5-HT6R antagonists. Design and synthesis of the series I (X = O, S; R1 = 4-PhOC6H4, 4-PhC6H4, 2-naphthyl, 1-naphthyl; R2 = H, Me, Et, n-Bu, R3 = H; R2 = R3 = Me) of ether and thioether derivatives of 1,3,5-triazines with the double-ring aromatic region have been performed. The new compounds I were examined within the comprehensive pharmacol. screening, including: radioligand binding assays, functional and ADMET studies in vitro as well as behavioral tests in rats. Crystallog. aspects and computer-aided structure-activity relationship were analyzed as well. This comprehensive approach led to selection of the compound I [X = S; R1 = 2-naphthyl; R2 = R3 = Me; (II)] with the most significant dual 5-HT6/5-HT2AR antagonistic action (5-HT6R Ki = 11 nM, 5-HT2AR Ki = 39 nM). Moreover, the compound II has satisfactory ADMETox properties in vitro, i.e.: the high permeability through biol. membranes, high metabolic stability, neither mutagenic nor hepatotoxic effects and moderate ability to inhibit CYP3A4. Above all, the compound II showed ability to reverse the pharmacol.-induced (MK-801) memory impairment at low doses (1-3 mg/kg) in Novel Object Recognition (NOR) test in rats. These results indicate a promising potency of dual 5-HT6/5-HT2AR antagonism in the search for novel strategy to fight Alzheimer’s disease, which remains an unmet clin. need.

A novel strategy for lead identification that we have dubbed the “Pocket-to-Lead” strategy is demonstrated using HIV-1 protease as a model target. Sometimes, it is difficult to obtain hit compounds because of the difficulties in satisfying the complex pharmacophoric features. In this study, a virtual fragment hit which does not match all of the pharmacophore features but has key interactions and vectors that could grow into remaining pharmacophore features was optimized in silico. The designed compound 9 demonstrated weak but evident inhibitory activity (IC50 = 54 μM), and the design concept was proven by the co-crystal structure. Then, structure-based drug design promptly gave compound 14 (IC50 = 0.0071 μM, EC50 = 0.86 μM), an almost 10,000-fold improvement in activity from 9. The structure of the designed mols. proved to be novel with high synthetic feasibility, indicating the usefulness of this strategy to tackle tough targets with complex pharmacophore.

Karpus, Andrii;Harrisson, Simon;Poli, Rinaldo;Mazieres, Stephane;Manoury, Eric;Destarac, Mathias research published 《 Well-Defined PIII-Terminated Polymers from Phosphorylated Carbodithioate RAFT Agents》, the research content is summarized as follows. Five S-alkyl di-tert-butylphosphorylated carbodithioates, Z-C(S)SR, were studied as chain transfer agents (CTAs) in the RAFT polymerization of vinyl monomers. These phosphorus-containing CTAs involve new PIII and PV moieties in the Z group (phosphine-borane or free phosphine). Two of them, with an S-bonded 1-methoxycarbonylethyl group as leaving group, have not previously been described. The CTAs with a PV Z group provide good control over the polymerization of styrene (St) and Bu acrylate (nBA). Molar masses are close to theor. values, and dispersities are consistently low during polymerization The monitoring of polymerization by 31P NMR spectroscopy supports the controlled character and the integrity of the polymer chain ends. Conversely, they inhibit the polymerization of the less-activated monomer vinyl acetate (VAc). The reactivity of the CTAs with a PIII Z group mainly depends on the reaction medium, and their performance for St, nBA, and VAc was surprising and unexpected. Theor. calculations were carried out to rationalize their behavior. Finally, an innovative strategy was developed to obtain well-controlled polymers with free phosphine ω-chain ends.
