A visible light-induced reductive addition of 1°, 2° and 3° alkyl iodides to ethenesulfonyl fluoride, employing Hantzsch ester as a hydrogen source at room temperature was developed for the synthesis of alkyl sulfonyl fluorides RCH2CH2SO2F [R = cyclohexyl, Bn, 1-adamantyl, etc.]. This method featured a wide substrate scope and great functional group compatibility, providing facile and robust process to alkyl sulfonyl fluorides including enzyme inhibitors, natural products and drugs derivatives in up to 99% yield. Further derivatization of resultant aliphatic sulfonyl fluorides was also achieved through sulfur fluoride exchange (SuFEx) reactions to deliver sulfonates and sulfonamides as privileged motifs for medicinal chem.

Zhang, Xuan; He, Yonghan;Zhang, Peiyi;Budamagunta, Vivekananda;Lv, Dongwen;Thummuri, Dinesh;Yang, Yang;Pei, Jing;Yuan, Yaxia;Zhou, Daohong;Zheng, Guangrong research published 《 Discovery of IAP-recruiting BCL-XL PROTACs as potent degraders across multiple cancer cell lines》, the research content is summarized as follows. Targeting BCL-XL via PROTACs is a promising strategy in reducing BCL-XL inhibition associated platelet toxicity. Recently, we reported potent BCL-XL PROTAC degraders that recruit VHL or CRBN E3 ligase. However, low protein expression or mutation of the responsible E3 ligase has been known to result in decreased protein degradation efficiency of the corresponding PROTACs. To overcome these mechanisms of resistance, PROTACs based on recruiting alternative E3 ligases could be generated. Thus, we designed and synthesized a series of PROTACs that recruit IAP E3 ligases for BCL-XL degradation Among those PROTACs, compound 8a(I) efficiently degrades BCL-XL in malignant T-cell lymphoma cell line MyLa 1929 while CRBN-based PROTACs that have high potency in other cancer cell lines show compromised potency, likely due to the low CRBN expression. Moreover, compared with the parent compound ABT-263, PROTAC 8a shows comparable cell killing effects in MyLa 1929 cells whereas the on-target platelet toxicity is significantly reduced. Our findings expand the anti-tumor spectra of BCL-XL degraders and further highlight the importance of selecting suitable E3 members to achieve effective cellular activity.

Twenty-nine 12 N-substituted aloperine derivatives were synthesized and screened for suppression on PD-L1 expression in H460 cells, as a continuation of our work. Systematic structural modifications led to the identification of compound I as the most active PD-L1 modulator. Compound I could significantly down-regulate both constitutive and inductive PD-L1 expression in NSCLC cells, and successively enhance the cytotoxicity of co-cultured T cells against tumor cells at the concentration of 20μM. Also, it exhibited a moderate in vivo anticancer efficacy against Lewis tumor xenograft with a stable PK and safety profile. The mechanism study indicated that I mediated the degradation of PD-L1 through a proteasome pathway, rather than a lysosome route. These results provided the powerful information for cancer immunotherapy of aloperine derivatives with unique endocyclic skeleton by targeting PD-L1 to activate immune cells to kill cancer cells.

Synthesis of pyrrolo[1,2-a]thiazolo[5,4-d]pyrimidinone derivatives I (R = Ph, 2-methylphenyl, 4-(trifluoromethyl)phenyl, etc.) were designed as cholinesterase inhibitor agents. The in vitro enzyme assays proved that most of the compounds effectively inhibited cholinesterases in the micromolar range with little cytotoxicity. Compound I (R = 4-(trifluoromethyl)phenyl) exhibited the best inhibitory activity against acetylcholinesterase with an IC50 of 2.69±0.17μ M. Kinetic anal. and mol. modeling testified that the competitive inhibition manner of I (R = 4-(trifluoromethyl)phenyl) was more likely to bind to the active center of acetylcholinesterase. Thus, compound I (R = 4-(trifluoromethyl)phenyl) can be considered as promising lead compounds or candidates for the development of novel drugs against Alzheimer’ disease.

A versatile and scalable strategy is reported for the rapid generation of block copolymer libraries spanning a wide range of compositions starting from a single parent copolymer. This strategy employs automated and operationally simple chromatog. separation that is demonstrated to be applicable to a variety of block copolymer chemistries on multigram scales with excellent mass recovery. The corresponding phase diagrams exhibit increased compositional resolution compared to those traditionally constructed via multiple, individual block copolymer syntheses. Increased uniformity and lower dispersity of the chromatog. libraries lead to differences in the location of order-order transitions and observable morphologies, highlighting the influence of dispersity on the self-assembly of block copolymers. Significantly, this separation technique greatly simplifies the exploration of block copolymer phase space across a range of compositions, monomer pairs, and mol. weights (up to 50000 amu), producing materials with increased control and homogeneity when compared to conventional strategies.

The radical mechanism is initiated by light absorption by the ruthenium complex at the center of the molecule which induces photoinduced electron transfer from the ruthenium to 1BDMB. This process results in electron transfer from the donor to an acceptor molecule, such as oxygen or nitrogen. The pharmacokinetic properties of this compound are not well known; however, it has been demonstrated that it can be synthesized through a cross-coupling reaction with other aromatic compounds such as stemofuran., Synthetic Route of 20469-65-2

Zhang, Dejiang;Tang, Ting;Zhang, Zhao;Le, Liyuan;Xu, Zhi;Lu, Hao;Tong, Zhou;Zeng, Dishu;Wong, Wai-Yeung;Yin, Shuang-Feng;Ghaderi, Arash;Kambe, Nobuaki;Qiu, Renhua research published 《 Nickel- and Palladium-Catalyzed Cross-Coupling of Stibines with Organic Halides: Site-Selective Sequential Reactions with Polyhalogenated Arenes》, the research content is summarized as follows. Herein, the authors disclose a general and efficient method for the synthesis of Sb-aryl and Sb-alkyl stibines by the Ni-catalyzed cross-coupling of halostibines with organic halides. The synthesized Sb-aryl stibines couple with aryl halides to give biaryls efficiently via Pd catalysis. Sequential reactions of stibines with polyhalogenated arenes bearing active C-I/C-Br sites and inactive C-Cl sites successfully proceeded, giving a variety of complex mols. with good site selectivity. Drugs such as diflunisal and fenbufen, as well as a fenofibrate derivative, were synthesized on gram scales in good yields, together with the high recovery of chlorostibine. Also, catalytic mechanisms are proposed based on the results of control experiments
