Mitochondria specific oxidative injury by near-infrared energy transfer nanoclusters for amplified photodynamic potency

To promote practical applications of photodynamic therapy, near-infrared (NIR) photosensitizers are manufactured based on fluorescence resonance energy transfer (FRET) between donors of anti-stoke NIR upconversion nanoparticles and acceptors of photodynamic chlorin e6. The manufactured FRET constructs displayed deep tissue penetration and FRET activation under 980 nm irradiation. Furthermore, surface decoration with mitochondria-targeting (4-marboxybutyl) triphenyl phosphonium bromide (TPP) led to mitochondrion-targeted accumulation of singlet oxygen resulting in potent cell apoptosis. Notably, in vivo anti-tumor test validates the complete ablation of intractable solid tumors based on single-dose treatment of our proposed photodynamic constructs. Therefore, the obtained results herald the tempting perspective of our carefully engineered photodynamic constructs, which could have broad utilities in clinical treatment of intractable premalignancies. Meltblown Polylactic Acid Nanowebs as a Tissue Engineering Scaffold

Polylactic acid (PLA) nanofiber nonwovens have recently come under more vigorous investigation for their use as tissue engineering scaffolds owing to its ability to mimic the physical properties of naturally occurring human extracellular matrix in a variety of host tissues. Currently, the majority of available research on PLA nanowebs has focused on their creation through electrospinning. The goal of this study was to evaluate meltblown nonwoven webs made of nanodiameter PLA fibers for their application as a tissue engineering scaffold. Meltblown PLA fabrics were produced with a variety of different crystallinities, tensile moduli, and pore diameters. One fabric with mechanical properties similar to human dermis was selected as a scaffold to study attachment, proliferation, and migration of human dermal fibroblasts over 1, 3, 7, and 14 days without the use of additional cell adhesion molecules. The 3-(4,5-dimethylthiazol-2-yl)-diphenyltetrazolium bromide assay showed good proliferation from day 1 to 3 (P = 0.026) and up to 7 days of culture (P = 0.005) but without increase from day 7 to 14. Electron microscopy demonstrated adequate cellular attachment and surface migration at 1, 3, 7, and 14 days. Finally, confocal microscopy was used to investigate cellular penetration into the scaffolds. The investigation found that cells were able to penetrate fully through the thickness of the scaffold. The successes of this initial experiment are promising and confirm that meltblown nanofiber nonwovens are a viable avenue for tissue engineering scaffolds. Hopefully, these conclusions will open the door for others to pursue research in this exciting field.

Porous nanosheets of carbon-conjugated graphitic carbon nitride for the oxidation of H2S to elemental sulfur

Graphitic carbon nitride (CN) has been reported as an emerging non-metallic catalyst for the oxidation of H2S to elemental sulfur, but its industrial application is hampered by the unsatisfactory catalytic activity. Herein, we report for the first time a facile bottom-up approach to synthesize porous nanosheets of in-plane carbon-conjugated CN (PCNUC) using cetyltrimethylammonium bromide (CTAB) and urea as precursors. The resulted PCNUC catalysts are porous and high in specific surface area because CTAB acts not only as a carbon source for conjugation with CN for the extension of pi-electron system, but also releases gases to function as soft template to create porous CN-based sheets. Thus, the carbon-conjugated CN displays high performance for the catalytic oxidation of H2S to S. Furthermore, the optimized PCNUC catalyst exhibits better durability than the traditional carbon-based and Fe2O3-based catalysts. The results of the present study open up a new avenue for the design of efficient CN-based catalysts for desulfurization reactions. (C) 2019 Published by Elsevier Ltd.

Evaluation of new chemical and biological nematicides for managing Meloidogyne javanica in tomato production and associated double-crops in Florida

BACKGROUNDTwo field experiments were conducted to determine the efficacy and field performance of three new non-fumigant chemical nematicides (fluensulfone, fluopyram, and fluazaindolizine) and two biological nematicides (Burkholderia rinojensis strain A396 and Purpureocillium lilacinus strain 251) for management of root-knot nematodes (Meloidogyne javanica) on tomato and associated double-crops in Florida. RESULTSIn experiment 1, soil fumigation with metam potassium increased plant growth and reduced root galling on tomato by 77% relative to that of the untreated soil. All non-fumigant chemical nematicides reduced root galling on tomato (47-85% reduction); however, only fluensulfone showed a trend towards yield enhancement. In experiment 2, soil fumigation with chloropicrin increased plant growth and reduced root galling on tomato by 35% relative to that of the untreated soil; however, end-of-season populations of M. javanica in soil were larger than that of the non-fumigated soil. Fluensulfone showed a trend towards reduced root galling and enhanced fruit yield, whereas other non-fumigant nematicides did not. Double-cropped cucumber was 69% more galled when planted into soil previously fumigated with chloropicrin relative to that of untreated soil, and also showed reduced plant vigor and fruit yield. Effect of beta-cyclodextrin, alcohol and temperature on the aggregation transitions of cationic/cationic surfactant systems

The mutual transitions between micelles and vesicles induced by beta-cyclodextrin (beta-CD), alcohol and temperatures, which had rarely been reported in mixed cationic/cationic surfactant systems, was studied in a dodecyltrimethylammonium chloride (DTAC)/didodecyldimethylammonium bromide (DDAB) aqueous solution. The effect of beta-CD, temperature and alcohol on this mixed system was investigated by the methods of turbidity, transmission electron microscopy (TEM), dynamic light scattering (DLS), and ‘H Nuclear magnetic resonance (H-1 NMR) measurements. We found that increasing the concentration of beta-CD would induce the micelle to vesicle (M-V) transition. While the vesicle to micelle (V-M) transition further occurred with increasing temperature or adding alcohol in this beta-CD/cationic/cationic mixed system. The M-V transition with adding beta-CD has the mechanism that the inclusion complexation of beta-CD to surfactants alters the composition ratio of the DDAB/DTAC systems, which changes the packing parameter. The V-M transition induced by temperature or alcohols is explained as the change of electrostatic repulsion interaction among the similar cationic polar heads in vesicles. The heat-induced V-M transition also verifies the M-V transition mechanism. Moreover, we firstly found that this heat-induced V-M transition was not only related to the percentage of DDAB concentration, but also to the total concentration of surfactant in cationic/cationic mixed surfactant systems.
