Cytoprotective effect of crocin and trans-resveratrol on photodamaged primary human retinal pigment epithelial cells

Purpose: Light-induced damage to retinal pigment epithelium during pars plana vitrectomy remains a hot topic in ophthalmology. Improvements in technology led to a change of light sources, selective filters, and shorter light exposure time. Currently, there is no satisfying solution to the problem. The aim of the study was to investigate the cytoprotective effects of crocin and resveratrol on light-induced damage to primary human retinal pigment epithelial cells in vitro. Methods: Primary human retinal pigment epithelial cells were exposed to light analogous to the illumination during pars plana vitrectomy. To evaluate the cytoprotective effects and potential toxicity of resveratrol and crocin, human retinal pigment epithelial cells were incubated with varying concentrations of both before 3-[4,5-dimethylthiazol-2-yl] tetrazolium bromide (MTT) viability assay. Furthermore, glutathione levels were measured to investigate synergistic antioxidant potential. Apoptosis of human retinal pigment epithelial cells was determined by a nucleosome detection enzyme-linked immunosorbent assay. Results: Crocin and resveratrol improved cell viability in photodamaged human retinal pigment epithelial cells significantly from 40.65 +/- 21.99% in illuminated human retinal pigment epithelial cells and reached a peak viability of 85.64 +/- 11.37% in crocin and resveratrol pretreated cells (for all: p < 0.001). In line, the combination of the supplements increased glutathione levels significantly from 39.35 +/- 21.96% to 80.74 +/- 10.32% (p = 0.017). No toxic effects were detected (p > 0.99). However, no change in apoptosis rates could be observed following pretreatment with crocin and resveratrol (p > 0.99). Conclusion: Crocin and trans-resveratrol revealed cytoprotective effects on human retinal pigment epithelial cells supporting both supplement’s development as potential perioperative treatments in light-induced retinal pigment epithelial damage.

Variability in care for children with severe acute asthma in Latin America

Background: Care variability for children with severe acute asthma has been well documented in high-income countries, yet data from low- and middle-income regions are lacking. We sought to characterize the magnitude of practice variability in the care of Latin American children to identify opportunities for standardization of care. Methods: A cross-sectional study performed through a retrospective analysis of contemporaneously collected data of children with severe acute asthma admitted to a center contributing to the LARed Network registry between May 2017 and May 2019. Centers were grouped by geographic location: Atlantic (AT), South Pacific (SP), and North Central (NC). Results: Among 434 children, most received care in hospitals in the AT group (54% [235/434]), followed by the NC (23% [101/434]) and SP (23% [98/434]) groups. The majority of children in the AT (92% [215/235]) and SP (91% [89/98]) groups received nebulized salbutamol/albuterol, while metered-dose inhalers were preferred in the NC group (72% [73/101]). There was a wide variation in the use of antibiotics: AT (57% [135/235]), SP (48% [47/98]), and NC (14% [14/101]). The same was true for ipratropium bromide: AT (67% [157/235]), SP (90% [88/98]), and NC (17% [17/101]), and aminophylline: AT (57% [135/235]), NC (5% [5/101]), and SP (0% [0/98]). High-flow nasal cannula was the preferred respiratory support modality in the AT (60% [141/235]) and NC (40% [40/101]) groups, while bilevel positive airway pressure (BiPAP) use was more common in the SP group (80% [78/98]). Conclusion: We identified significant variability in care for severe acute asthma. Our findings will help to inform the design of future studies, quality improvement initiatives, and development of practice guidelines within Latin America.

Adenosine 5’-triphosphate (ATP), as a molecular currency for intracellular energy transfer, not only participates in various physiological activities, but also involves in various pathological processes of living cells. Therefore, the establishment of a simple, rapid, sensitive and accurate ATP analysis method is of great significance for the study of its physiological functions. In this work, we rationally design a near-infrared phenylboronic acid group-functionalized dicyanovinyl squaraine (SQ- PBA3), with which a supramolecular assembly was constructed for fluorescent imaging of ATP. In PBS buffer, SQ-PBA3 changes self-assembly when adding cetyltrimethyl ammonium bromide (CTAB) deriving fluorescence response of SQ-PBA3 to ATP with the maximum emission at 700 nm, due to the specificity of phenylboronic acid to diols and the multiple electrostatic interactions between SQ PBA3, ATP and CTAB molecules. Meanwhile, the probe was successfully applied to monitor intracellular ATP level in MCF-7 cells with high sensitivity and selectivity.

Effects of silica nanoparticle on the solution properties of hydrophobically associating polymer based on acrylamide and beta-cyclodextrin

Synthesis of a novel beta-cyclodextrin-functionalized hydrophobically associating polyacrylamide named ASM was carried out using acrylamide (AM), cetyl dimethyl allyl ammonium bromide (SD-16) and modified beta-cyclodextrin (M-beta-CD) via free-radical copolymerization. Effective characterization methods, including FT-IR, H-1 NMR and TGA, were adopted to evaluate ASM. Influences of SiO2 nanoparticle (nanoSiO(2)) on ASM solution properties were evaluated. The results showed that the addition of nano-SiO2 significantly increased the temperature resistance, salt tolerance and shear resistance of the ASM solution. It was found that ASM/SiO2 composite had 1.5 times higher viscosity than ASM in the range of 25 -90 degrees C. The FT-IR data confirmed that the formation of hydrogen bond between silanol groups at surface of nano-SiO2 and amide group, hydroxyl group of M-beta-CD in ASM molecule contributed to the improved performance. What s more, viscoelastic test indicated that nano-SiO2 remarkably facilitated the crosslinks among polymer molecules and enhanced the elasticity of ASM/SiO2 composite. Materials and Methods: The effects of Sargassum miyabei Yendo extract (SME) on 3T3-L1 adipocytes were screened by3-(4,5)-dimethylthiazo-2-yl-2,5-diphenyltetrazolium bromide (MTT), Oil red O staining, western blot, and Real-time reverse transcription polymerase chain reaction analyses. Results: Here, we show that SME had potent 2,2′-azinobis-3-ehtlbezothiazoline-6-sulfonic acid radical decolorization (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) antioxidant activity with half maximal inhibitory concentration (IC50) value of 0.2868 +/- 0.011 mg/mL and 0.2941 +/- 0.014 mg/mL, respectively. In addition, SME significantly suppressed lipid accumulation and differentiation of 3T3-L1 preadipocytes, as shown by Oil Red O staining results. SME attenuated the expression of adipogenic- and lipogenic-related genes such as peroxisome proliferator-activated receptor gamma (PPAR gamma), CCAAT-enhancer-binding protein alpha (C/EBP alpha), CCAAT-enhancer-binding protein delta (C/EBP delta), adiponectin, adipose triglyceride lipase (ATGL), fatty acid synthase (FAS), hormone-sensitive lipase (HSL), and lipoprotein lipase (LPL). Conclusions: These findings suggest that SME may have therapeutic implications for developing a new anti-obesity agent.

Characteristics of zeolite ZSM-12 crystallization using methyltriethylammonium chloride and N,N-dimethyl-N-ethyl(monoethanol)ammonium bromide as templates have been studied. The materials have been characterized by X-ray diffraction analysis, scanning electron microscopy, X-ray fluorescence analysis, low-temperature nitrogen adsorption-desorption, temperature-programmed desorption of ammonia, and solid-state Al-27 nuclear magnetic resonance spectroscopy. It has been shown that dimethylethylethanolammonium bromide inhibits crystal growth along the a and c planes and thereby contributes to crystal growth only in the direction of the b axis, which, in turn, leads to lower surface area and acidity of the sample than the respective parameters of the zeolite synthesized using methyltriethylammonium chloride.

Magnetic Fe3O4 @ porous activated carbon effervescent tablet-assisted deep eutectic solvent-based dispersive liquid-liquid microextraction of phenolic endocrine disrupting chemicals in environmental water

A novel approach using magnetic Fe3O4 @ porous activated carbon effervescent tablet-assisted deep eutectic solvent-based dispersive liquid-liquid microextraction (META-DES-DLLME) combined with HPLC was developed for the measurement of phenolic endocrine disrupting chemicals (bisphenol A, p-tert-octylphenol, nonylphenol) in environmental water. In the proposed method, deep eutectic solvents synthesized by 1-dodecanol and hexyltrimethylammonium bromide were applied as extraction solvent. A magnetic effervescent tablet, containing a foaming agent and a new magnetic material, combined the dispersion of the extraction solvent and the magnetic recovery into one step. The influence of parameters, including the composition and volume of DESs, the type and amount of acid, the amount of sodium bicarbonate, the composition and amount of Fe3O4 @ porous activated carbon, the pH, and the amount of sodium chloride, were optimized. Under the optimized conditions, accuracy and precision were demonstrated over the whole concentration range of 10-1000 mu g L-1. The extraction recovery ranged from 81.0 to 94.7% with a relative standard deviation of 1.2-6.7%, and the LOD and LOQ ranged from 0.82 to 1.7 mu g L-1 and 2.7-5.5 mu g L-1, respectively. The method was successfully applied to extract and analyse the phenolic endocrine disrupting chemicals in environmental water.

The crystallization, thermal and mechanical properties of nano-Sb2O3 filled poly(butylene terephthalate)

Nano-Sb2O3 particles were modified by a combination modifier of cetyltrimethyl ammonium bromide (CTAB) and KH-560 via the mechanochemical method based on high-energy ball milling. Then, the testing specimens of the nano-Sb2O3/PBT composites of differing compositions were prepared by melting blending technology. The crystallization, thermal, and mechanical properties of composites were characterized by X-ray diffraction, differential scanning calorimetry, thermogravimetric analyzer, and mechanical performance test. The tensile and impact fracture surfaces of composites were determined by scanning electron microscopy. Besides, the influence of the Sb2O3 nanoparticles surface modification on crystallinity, mechanical properties of the composites, and the interfacial adhesion between nano-Sb2O3 and PBT was systematically investigated. The results indicate that the main crystalline characteristics of PBT matrix remain unchanged in the nanocomposites. However, the addition of nano-Sb2O3 particles plays a heterogeneous nucleation and can effectively improve the crystallization of PBT matrix. In addition, the compound modification of the nano-Sb2O3 can effectively enhance mechanical properties of the composites and interfacial interaction between nano-Sb2O3 and PBT. The enhanced fracture properties in the nanocomposites were caused by the assisted void formation at the edge of the nano-Sb2O3 particle. When the nano-Sb2O3 mass fraction is 3%, the composites show excellent comprehensive performance. The interfacial adhesion parameter B and the half-debonding angle theta of composites were assessed to quantitatively characterize the interfacial adhesion strength between nano-Sb2O3 and PBT.
