Surfactant-aided synthesis of RhCo nanoclusters as highly effective and recyclable catalysts for the hydrolysis of methylamine borane and dimethylamine borane

In the present study, the first ever and easy surfactant-aided synthesis of hexadecyltrimethyl ammonium bromide-stabilized RhCo nanoclusters (Rh0.63Co0.37@CTAB) and their employment as highly efficient, relatively cost-saving, and recyclable catalysts to generate hydrogen from the hydrolysis reactions of methylamine borane (MeAB) and dimethylamine borane (DMAB) are reported. The easy synthesis of Rh0.63Co0.37@CTAB nanoclusters is accomplished by synchronous reduction of appropriate rhodium and cobalt salts by sodium borohydride in the presence of CTAB as a surfactant in aqueous solution at room temperature. The Rh0.63Co0.37@CTAB nanoclusters are characterized by some techniques like ultraviolet-visible electronic absorption spectroscopy (UV-vis), transmission electron microscopy (TEM), and X-ray photoelectron spectroscopy (XPS). They provide very remarkable turnover frequencies (TOFs) of 107.1 min(-1) and 142.9 min(-1) (8571.4 h(-1), best ever value) in the hydrolysis of MeAB and DMAB, respectively, even at very low temperatures. Their activation energies in the same reactions are calculated to be 55.3 kJ mol(-1) and 50.3 kJ mol(-1), respectively.

Dispersing graphene in aqueous media: Investigating the effect of different surfactants

Performance of graphene nanoplatelets depends on its dispersion quality and level of homogeneity, to this end investigating graphene dispersing approaches from different point of view will be highly applicable. In this study, we compared the effects of different surfactants-sodium dodecylsulfate (SDS), sodium dodecylbenzen sulfonate (SDBS), cetyltrimethylammonium bromide (CTAB), and nonylphenolethoxylate (NPE)-in their capacity to produce stable aqueous media containing homogeneously dispersed graphene nanoplatelets (GNPs). To compare the surfactants’ ability to produce optimal dispersion of graphene, we relied on optical characterization, i.e. UV-vis spectroscopy, optical microscopy, and turbidimetry. CTAB-containing GNP dispersion showed the highest stability and lowest graphene flake size. The character of the different surfactants led us to investigate the stability mechanisms in the aqueous dispersions. Zeta potential was measured to determine the effective surface charge of GNPs in each aqueous medium. The accordance between experimental results and theories in this regard increases the reliability of results. Hence, we evaluated the results through DLVO theory to rank the performance of surfactants more evidently. Based on zeta potential values and the height of the energy barrier in interaction energy graph, CTAB had the best performance in terms of stability and preventing GNP aggregate formation. Molecular dynamics simulation was conducted to have better understanding of interactions between different surfactants and graphene nanosheets. The performance of the surfactants was ranked in order of CTAB, SDBS, SDS, NPE. Using UV-vis spectroscopy, particle size analysis, TEM and SEM imagery, we determined that the optimal concentration of CTAB is 400 ppm.

Interleukin-8 gene expression and apoptosis induced by Salmonella Typhimurium in the presence of Bacillus probiotics in the epithelial cell

Aims This study aimed to evaluate the effects of three Bacillus probiotics on Salmonella Typhimurium, and interleukin-8 (IL-8) gene expression in the co-culture of the Bacillus and the pathogen in vitro. Methods and Results Bacillus subtilis, Bacillus indicus and Bacillus coagulans were initially turned to spore and heat-inactivated forms. The cellular damages of the probiotics on the HT-29 cells were investigated individually and in combination with S. Typhimurium using 3-(4,5 dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) and fluorescence assays. To extract cell free supernatants (CFS) of the probiotics, they were cultured in selective media. The inhibitory activity of CFSs were then assayed against the pathogen. The gene expression of IL-8 of the HT-29 cells was evaluated by real-time PCR in all the groups. The results showed that the CFSs of three probiotics could inhibit the growth of S. Typhimurium by more than 50%. Inhibitory effects of B. indicus and B. subtilis CFSs were related to the production of pepsin-sensitive compounds, except B. coagulans in which the high inhibitory effect was due to organic acids. The spores of the three probiotics and the heat-inactivated forms of B. subtilis and B. coagulans could reduce the cytotoxicity of S. Typhimurium. The cell viability also increased applying both forms probiotics against the pathogen. In all co-culture groups, the IL-8 gene expression induced by S. Typhimurium was reduced. Conclusions The three Bacillus probiotics can be considered as proper candidates for the prevention and treatment of S. Typhimurium food poisoning. Significance and Impact of the Study Applying probiotics as live bacteria is universally noted in foods. This study tried to discover the effects of Bacillus probiotics in the form of spore or even heat-killed bacteria against S. Typhimurium and evaluate ratio of IL-8 gene expression in cell culture. Colorimetric Aptasensor for Testosterone Detection Based on Aggregation of Gold Nanoparticles Induced by Cationic Surfactant

This paper proposes a colorimetric aptasensor for the detection of testosterone (TES) in environmental water, using TES-specific aptamer (apT5) as a sensing probe, gold nanoparticles (AuNPs) as indicator, and hexadecyltrimethylammonium bromide (CTAB) as inducer, respectively. Based on competition between TES and CTAB for apT5, the aptamer can form an aptamer-TES complex, leaving CTAB free to aggregate AuNPs in the presence of TES. Dispersed and aggregated AuNPs have different absorption wavelengths and the signal of absorption intensity is associated with the concentration of TES, so TES can be detected quantitatively based on the signal absorption intensity. This sensitive aptasensor for TES detection has a wide linear range (R = 0.998) from 1.91-800nM and a limit of detection (LOD) of 1.91 nM. In addition, this aptasensor has high selectivity over some interferents. The method detects TES in tap water samples with recoveries in the range of 98.9-102.6%(RSD <= 7.35 %). Hydrogen sulfide detection and sensing is an area of interest from both an environmental and a biological perspective. While many methods are currently available, the most sensitive and biologically applicable ones are fluorescence based. In general, these fluorescent probes are based upon large, high-molecular weight, well-characterized fluorescent scaffolds that are synthetically demanding to prepare and difficult to tune and modify. In this study, we have reported a new reduction-based, rationally designed and synthesized turn-on fluorescent probe (Z)-2-(4 ‘-azidobenzylidene)-5-fluorobenzofuran-3(2H)-one (6g) utilizing a low molecular weight aurone fluorophore. During these studies, the modular nature of the synthesis was used to quickly overcome problems with solubility, overlap of excitation of the probe and reduced product, and rate of reaction, resulting in a final compound that is efficient and sensitive for the detection of hydrogen sulfide. The limitation of slow reaction and the reduced fluorescence in a biologically relevent medium was solved by employing cationic surfactant cetyltrimethyl ammonium bromide (CTAB). The probe features a high fluorescence enhancement, fast response (10-30 min), and good sensitivity (1 mu m) and selectivity for hydrogen sulfide.

Design, synthesis and biological evaluation of iridium(III) complexes as potential antitumor agents

Cisplatin and its analogs have been used for the treatment of various cancers, but their serious side effect has limited clinical application. Presently, scientists are developing other metal drugs as an alternative of cisplatin. In this paper, three new iridium(III) complexes [Ir(ppy)(2)(adppz)](PF6) (adppz = 7-aminodipyrido[3,2-alpha:2′,3′-c] phenazine; ppy = 2-phenylpyridine 1), [Ir(bzq)(2)(adppz)](PF6) (bzq = benzo[h]quinolone 2) and [Ir(piq)(2)(adppz)](PF6) (piq = 1-phenylisoquinoline 3) were synthesized and characterized. The complexes can effectively inhibit the cell colonies. The cytotoxicity in vitro of the complexes against A549, HepG2, SGC-7901, BEL-7402 and normal NIH3T3 cells was evaluated by 3-(4,5-dimethylthiazole)-2,5-diphenyltetraazolium bromide (MTT) methods. The intracellular reactive oxygen species (ROS) levels and Ca2+ concentrations were assayed. The mitochondrial membrane potential, a release of cytochrome c and the expression of B-cell lymphoma/leukemia-2 (Bcl-2) family protein have been investigated. The data reveal that the complexes 1-3 can effectively inhibit the cell proliferation in A549 cells with low IC50 value of 3.2 +/- 0.4 mu M, 4.8 +/- 0.5 and 1.2 +/- 0.2 mu M, respectively. The antitumor in vivo shows that complex 3 can inhibit tumor growth with an inhibitory rate of 76.34%. The studies on the mechanism indicate that these complexes cause apoptosis in A549 cell via a ROS-mediated lysosomal-mitochondrial dysfunction pathway. In addition, the interaction of the complexes with BSA was explored.

Nano-sized formazan analogues: Synthesis, structure elucidation, antimicrobial activity and docking study for COVID-19

Three series of nanosized-formazan analogues were synthesized from the reaction of dithiazone with various types of alpha-haloketones (ester and acetyl substituted hydrazonoyl chlorides and phenacyl bromides) in sodium ethoxide solution. The structure and the crystal size of the new synthesized derivatives were assured based on the spectral analyses, XRD and SEM data. The antibacterial and antifungal activities were evaluated by agar diffusion technique. The results showed mild to moderate antibacterial activities and moderate to potent antifungal activities. Significant antifungal activities were observed for four derivatives 3a, 3d, 5a and 5g on the pathogenic fungal strains; Aspergillus flavus and Candida albicans with inhibition zone ranging from 16 to 20 mm. Molecular docking simulations of the synthesized compounds into leucyl-tRNA synthetase editing domain of Candida albicans suggested that most formazan analogues can fit deeply forming stable complexes in the active site. Furthermore, we utilized the docking approach to examine the potential of these compounds to inhibit SARS-CoV-2 3CLpro. The results were very promising verifying these formazan analogues as a hopeful antiviral agents.

Voltammetric determination of prifinium bromide in a pharmaceutical formulation

For the first time voltammetric methods have been applied and optimized for prifinium bromide (PRF) determination in pharmaceutical formulation. Cyclic voltammetry (CV) study has shown that PRF is an electroactive compound with quasi-revisable voltammograms. Exosomes Derived from Bovine Mammary Epithelial Cells Treated with Transforming Growth Factor-beta 1 Inhibit the Proliferation of Bovine Macrophages

Transforming growth factor (TGF)-beta 1 is a multifunctional cytokine that plays an important role in regulating immune cell proliferation. We speculate that high expression of TGF-beta 1 may affect the immunity of dairy cows. In this study, untreated exosomes (un-exo) derived from an untreated bovine mammary epithelial cell line (MAC-T) and TGF-beta 1-treated exosomes (t-exo) derived from TGF-beta 1-treated MAC-T cells were isolated by ultracentrifugation and identified by electron microscopy and Western blotting. Then, un-exo and t-exo were used to treat a bovine macrophage cell line (BOSMAC), and the proliferative ability of BOSMAC cells was detected by an 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide assay and flow cytometry. The expression and phosphorylation levels of p38 were analyzed by q-PCR and Western blotting. The results showed that both exosome types exhibited the basic characteristics of exosomes. In BOSMAC cells treated with t-exo, significant inhibition of cell proliferation was observed, and the cell cycle progression was inhibited, while no difference was found between the un-exo and control groups. Only treatment with t-exo increased the expression and phosphorylation of p38, and the addition of the p38 inhibitor SB203580 abrogated the inhibition of BOSMAC cell proliferation by t-exo. Our results demonstrated that t-exo inhibited the proliferation of bovine macrophages by stimulating p38 MAPK and might interfere with immunity in dairy cattle. This finding may provide a new strategy for improving immunity and preventing breast-related diseases in dairy cows.
