Mitochondrial dysfunction is involved in aristolochic acid I-induced apoptosis in renal proximal tubular epithelial cells

Aristolochic acid (AA) is a compound extracted from the Aristolochia species of herbs. AA exposure is associated with kidney injury known as aristolochic acid nephropathy (AAN). Proximal tubular epithelial cell (PTEC) is the primary target of AA and rich in mitochondria. Recently, increasing evidence suggests that mitochondrial dysfunction plays a critical role in the pathogenesis of kidney disease. However, the status of mitochondrial function in PTEC after exposure to AA remains largely unknown. The aim of this study was to explore the effect of aristolochic acid I (AAI) on cell apoptosis and mitochondrial function in PTEC. Normal rat kidney-52E (NRK-52E) cells were exposed to different concentrations of AAI for different time periods. Cell viability was detected by 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide, cell apoptosis was analyzed by flow cytometry, and the expression of cleaved caspase-3 by Western blotting. Mitochondrial function was evaluated by reactive oxygen species (ROS), mitochondrial membrane potential (MMP), mitochondrial DNA (mtDNA) copy number, and adenosine triphosphate (ATP). It was found that AAI reduced cell viability and increased cell apoptosis in a dose- and time-dependent manner. In parallel to increased apoptosis, NRK-52E cell manifested signs of mitochondrial dysfunction in response to AAI treatment. The data indicated that AAI could increase ROS level, lower MMP, decrease mtDNA copy number, and reduce ATP production. In addition, Szeto-Schiller 31, a mitochondria-targeted antioxidant peptide, attenuated AAI-induced mitochondrial dysfunction and apoptosis. Our study depicted significant aberrant of mitochondrial function in AAI-treated NRK-52E cell, which suggested that mitochondrial dysfunction may be involved in AAI-induced apoptosis in PTEC.

Combinational therapy is among the most used treatment modality’s to increase cancer treatment efficacy. It may also reduce side effects, treatment time, and development of drug resistance. The effects of different analogues of vitamin A (VA) as neoadjuvant agent were observed in this study with chemotherapeutic drugs (doxorubicin and methotrexate) and photodynamic therapy (PDT) using 5-ALA and Photogem as photosensitizers in RD cells. The uptake time of photosensitizer was optimized by means of spectrophotometric measurements. Diode laser (lambda = 635 nm +/- 1 nm) was used as an illumination source for PDT. Responses of administered drugs were assessed by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. VA and its analogues exert prophylactic and therapeutic effects. Protective or antagonistic effects (CI > 1) were observed in each of the case. These results propose that the use of VA as a neoadjuvant agent in combinational therapeutic modalities may reduce the efficacy of cancer treatment protocols as well as the existing side effects. The stability and toxicity of TriCurin microemulsion were tested in an organotypic cervical tissue model. Results TriCurin and its individual compounds inhibit the growth of W12 cells, episomal, type 1 and 2 integrants; the relative order of activity is TriCurin, EGCG, curcumin, or resveratrol. RT-PCR shows that TriCurin activates p53 and suppresses HPV16 mRNAs E1, E2, E4, E6 and E7 at 24 h in W12 cells. Gene expression analysis shows that TriCurin activates pro-apoptotic genes and represses anti-apoptotic genes in HeLa cells. TriCurin in a microemulsion is stable and non-toxic to cervical tissue. Herein we report the first example of an on-water enantiospecific synthesis of alkyl bromides. Repellence and acute toxicity of a nano-emulsion of sweet orange essential oil toward two major stored grain insect pests

Control strategies in stored-product facilities mainly rely on synthetic pesticides. The development of new environmentally friendly alternatives, such as essential oils (EOs), is a key issue. The aim of this research was to develop a stable nano-emulsion containing a high amount of sweet orange (Citrus sinensis) EO and to evaluate its insecticidal activity against Tribolium confusum du Val (Coloptera: Tenebrionidae) and Cryptolestes ferrugineus (Stephens) (Coleoptera: Cucujidae). The experimental nano-emulsion showed a low surface charge (zeta slightly higher than -30 mV) and a droplet size within the sub-micrometre range (131.37 nm +/- 0.29), maintained also after 1 year of storage. The EO nano-emulsion presented good repellence toward both adult insects, which lasted until 24 h of exposure at the highest dosages. Furthermore, the developed nano-formulation showed acute toxicity against both insects when tested as fumigant and cold aerosol. In fumigation trials, adults of C. ferrugineus were more susceptible than T. confusum adults. Moreover, the insecticidal activity of cold aerosol was 5 and 7-fold higher than fumigation against both C. ferrugineus and T. confusum, respectively. The EO-nano-emulsion was effective in controlling and repelling the target pests. Cold aerosol treatments with EO nano-formulations is a promising alternative method for the sanitation of production areas, warehouses, and machineries.

Glucose-Derived N-Doped Graphitic Carbon: Facile One-Pot Graphitic Structure-Controlled Chemical Synthesis with Comprehensive Insight into the Controlling Mechanisms

Modification of graphitic structure chemistry and physics affords the N-doped graphitic carbonaceous materials (NGCMs) superior and novel functional properties, which is essential for substantial development of applications based on such materials. The present research provides novel simple structure-controlled chemical synthesis of NGCM. NGCM is grown from cyclic polymerization of glucose molecules upon hydrothermal treatment, employing Cetyltrimethylammonium bromide (CTAB) and ammonia as structure-directing agents. Hydrothermal temperature and CTAB dose were manipulated and the produced graphitic structure has been explored in terms of oxidation state, N-doping, surface functionalization with CTAB and morphology. NGC derivatives of various structural features are obtained. Products range from oxidized graphitic carbon of mixed micro-sized sphere/sheet morphology to highly-reduced graphene-like nanosheets; the C/O atomic ratio increases from 8.2-12.4 and sheet thickness downs from 30 mu m- similar to 1 nm. Also, N-doping configurations and graphitic surface functionalization by CTAB are greatly influenced. CTAB plays the key role in setting the graphitic structure characteristics. Additionally, ammonia has a modest effect. Understanding of the controlling mechanisms is focused. The effective and ease control of graphitic structure via this facile, economic and ecofriendly synthesis approach, can benefit the design of NGCM with tuned functional properties for specific applications in technological industries.
